Test Cross

Eine Testkreuzung ist eine Technik, die zur Bestimmung des Genotyps eines Organismus, der ein dominantes Merkmal zeigt, verwendet werden können.
 Zum Beispiel eine violettblütige Erbsenpflanze.
 Weil die dominante Version oder das Allel eines Gens immer über das rezessive Allel ausgedrückt wird, sehen Organismen mit dem homozygoten-dominanten Genotyp und zwei dominanten Allele gleich aus wie Organismen mit dem heterozygoten Genotyp, einem dominanten Allel.

 Und man weiß, dass sie den homozygot-rezessive Genotyp hat, klein p, klein p.
 In einem Szenario, in dem alle Nachkommen aus der Testkreuzung den dominanten Phänotyp zeigen, muss das unbekannte Elternteil homozygot-dominant für violette Blüten sein.

 Ein Allel ist rezessiv, wenn seine Wirkungen durch ein anderes Allel am gleichen Genlocus verdeckt werden. Zum Beispiel können Erbsenpflanzen violette oder weiße Blüten haben. In diesem Beispiel sind weiße Blüten rezessiv, da eine einzelne Kopie des Allels für violette Blüten zu einer Pflanze mit violetten Blüten führt, auch wenn diese Pflanzen das Allel für weiße Blüten haben.
 
 Wenn ein Organismus ein bekanntes rezessives Merkmal aufweist, ist die Bestimmung des Genotyps des Organismus und des Paares der vererbten Allele einfach. Nur ein Genotyp, pp (zwei rezessive Allele), produziert weiße Blüten.

 Wissenschaftler führen Testkreuzungen durch, um die Genotypen von Organismen zu bestimmen, die einfache dominante Merkmale aufweisen. Bei einer Testkreuzung wird der zu bestimmende Organismus mit einem Organismus gezüchtet oder gekreuzt, der das rezessive Gegenstück des dominanten Merkmals aufweist.
 
 Bei einer Testkreuzung für eine Erbsenpflanze mit violetten Blüten wird mit einer Erbsenpflanze mit weißen Blüten (pp-Genotyp) gekreuzt. Die Blütenfarbe der entstehenden Nachkommen zeigt, ob der Elternteil mit violetten Blüten homozygot (PP) oder heterozygot (Pp) ist.
 
 Wenn die Pflanze homozygot ist, erben alle Nachkommen eines ihrer dominanten P-Allele und eines der anderen rezessiven P-Allele der Eltern. Da das dominante Allel das rezessive Allele verdeckt, haben alle Nachkommen violette Blüten, das dominante Merkmal.
 
 Wenn die Pflanze jedoch heterozygot ist, erben etwa 50% der Nachkommen ihr rezessives p-Allel, zusammen mit einem weiteren rezessiven Allel von dem anderen Elternteil. Daher wird etwa die Hälfte der Nachkommen aus dieser Kreuzung weiße Blüten haben. Auf diese Weise kann man durch Testkreuzungen unbekannte elterliche Genotypen bestimmen.

Test crosses are used to identify the genotype of an organism with a dominant trait. A genotype is the pair of alleles inherited for a gene, and alleles are different forms of the same gene. In diploid organisms, including humans, each gene usually has two alleles, one from each parent.

An allele is recessive when its effect is hidden by another allele at the same gene location. Pea plants provide a clear example with flower color. White flowers are recessive, because one copy of the purple allele is enough to make a plant purple, even if it also carries the white allele.

When a trait is recessive, the genotype is easy to determine. For white pea flowers, only one genotype produces the trait: pp, with two recessive alleles. The problem is different for dominant traits, such as purple flowers. Two genotypes, PP and Pp, can both produce purple flowers.

Scientists use a test cross to solve that problem. In this method, the organism with the dominant trait is bred with an organism that shows the recessive version of the same trait. For a purple-flowered pea plant, the cross is made with a white-flowered pea plant with the pp genotype. The offspring then show whether the purple parent is homozygous, meaning PP, or heterozygous, meaning Pp.

The offspring pattern gives the answer. If the purple plant is homozygous, every offspring inherits one dominant P allele and one recessive p allele, so they all have purple flowers. If the purple plant is heterozygous, about half of the offspring inherit the recessive p allele from that parent and become white. In this way, test crosses reveal unknown parental genotypes.
