Die DNA-Replikation erfolgt durch die Synthese neuer DNA-Stränge unter Verwendung vorhandener Stränge als Vorlage. Die beiden neuen Doppelhelices enthalten jeweils einen ursprünglichen Template-Strang und einen neu synthetisierten Tochterstrang, weshalb dieser Prozess als semikonservative Replikation bezeichnet wird. 
Um die Replikation zu starten, wickelt ein Enzym, die Helikase, die DNA-Helix ab und bricht die Wasserstoffbrückenbindungen zwischen den beiden Strängen auf. Anschließend trennt es die einzelnen Stränge zu einer Y-förmigen Struktur, der sogenannten Replikationsgabel, auf die weitere Enzyme zugreifen können.
Ein anderes Enzym, die Primase, fügt jedem Matrizenstrang kurze RNA-Fragmente, sogenannte Primer, hinzu. Diese Primer sind für die Synthese von DNA unerlässlich, da die DNA-Polymerase nur Nukleotide an einen vorhandenen Strang anhängen kann. 
DNA-Polymerase fügt sich zu den wachsenden Tochtersträngen auf beiden Matrizen-DNA-Strängen hinzu. Die Zugabe von Nukleotiden richtet sich nach der Sequenz des ursprünglichen DNA-Strangs gemäß den Regeln der DNA-Paarung.
Die Synthese eines der Tochterstränge, des führenden Strangs, erfolgt in Richtung der Bewegung der Replikationsgabel. Der andere Strang, der nacheilende Strang, wird in die entgegengesetzte Richtung synthetisiert.
Dies führt dazu, dass der führende Strang als kontinuierliches Polymer synthetisiert wird, während der nacheilende Strang in Form von kurzen Fragmenten synthetisiert wird. Dies liegt daran, dass DNA nur in 5'- bis 3'-Richtung synthetisiert werden kann.
Vor ihrer Zugabe an das wachsende Polymer liegen Nukleotide als Desoxyribonukleosidtriphosphate vor, wobei drei Phosphate an den fünften Kohlenstoff auf dem Zucker gebunden sind.
Dieses freie Nukleotidtriphosphat reagiert mit den 3'-Hydroxylgruppen. Dies ist das OH, das am Ende des Wachstumsstrangs an den dritten Kohlenstoff des Zuckers gebunden ist. Die Reaktion bewirkt die Freisetzung von Pyrophosphat und die Bildung einer Phosphodiesterbindung zwischen den beiden Nukleotiden.
Nach der Synthese der neuen Stränge entfernen RNase H oder weitere Varianten der DNA-Polymerase die Primer und synthetisieren an ihrer Stelle DNA. Die Lücken zwischen den Fragmenten werden dann durch DNA-Ligase versiegelt, um einen durchgehenden Strang zu erzeugen.
Die Zugabe von Nukleotiden wird so lange fortgesetzt, bis zwei Replikationsgabeln aufeinandertreffen, was zur abgeschlossenen Replikation führt.

DNA Replication

Bei der DNA-Replikation werden die beiden Stränge der Doppelhelix getrennt, wobei jeder Strang als Vorlage dient, von der der neue komplementäre Strang kopiert wird. Nach der Replikation enthält jede doppelsträngige DNA einen elterlichen- oder „alten“ Strang und einen „neuen“ Strang. Dies wird als semikonservative Replikation bezeichnet. Die resultierenden DNA-Moleküle haben die gleiche Sequenz und werden gleichmäßig auf die beiden Tochterzellen aufgeteilt. Replikation in Prokaryoten Die DNA-Replikation nutzt eine große Anzahl von Proteinen und Enzymen. Das Enzym Helikase trennt die beiden DNA-Stränge. Während sich die Helikase entlang der DNA bewegt, trennt sie die beiden Stränge und bildet eine Y-förmige Struktur, die als Replikationsgabel bekannt ist. Anschließend fügt das Enzym Primase kurze RNA-Abschnitte, sogenannte Primer, hinzu, um die DNA-Synthese durch die DNA-Polymerase, das für die DNA-Synthese verantwortliche Enzym, zu initiieren. In Bakterien sind drei Haupttypen von DNA-Polymerasen bekannt: DNA pol I, DNA pol II und DNA pol III. DNA pol III ist das für die DNA-Synthese erforderliche Enzym; Für die Reparatur werden vor allem DNA pol I und DNA pol II benötigt. DNA pol III fügt der 3’-OH-Gruppe der wachsenden DNA-Kette Desoxyribonukleotide hinzu, die jeweils zu einem Nukleotid auf dem Matrizenstrang komplementär sind. DNA-Polymerase III kann sich nur in der 5’- bis 3’-Richtung ausdehnen. Die Zugabe dieser Nukleotide erfordert Energie. Diese Energie ist in den Bindungen von drei Phosphatgruppen vorhanden, die an jedes Nukleotid gebunden sind, ähnlich wie Energie in den Phosphatbindungen von Adenosintriphosphat (ATP) gespeichert wird. Wenn die Bindung zwischen den Phosphaten aufgebrochen wird und Diphosphat freigesetzt wird, ermöglicht die freigesetzte Energie die Bildung einer kovalenten Phosphodiesterbindung durch Dehydratisierungssynthese. Die DNA-Doppelhelix ist antiparallel; Das heißt, ein Strang ist in der 5’-zu-3’-Richtung und der andere in der 3’-zu-5’-Richtung ausgerichtet. Während der Replikation wird ein Strang, der zum 3‘- bis 5‘-elterlichen-DNA-Strang komplementär ist, kontinuierlich in Richtung der Replikationsgabel synthetisiert, da die Polymerase in dieser Richtung Nukleotide hinzufügen kann. Dieser kontinuierlich synthetisierte Strang wird als Leitstrang bezeichnet. Der andere Strang, der zur 5'- bis 3'-elterlichen-DNA komplementär ist, wächst von der Replikationsgabel weg, daher muss sich die Polymerase zurück zur Replikationsgabel bewegen, um mit dem Hinzufügen von Basen zu einem neuen Primer zu beginnen, wiederum in der Richtung weg von der Replikationsgabel. Dies geschieht so lange, bis es auf den zuvor synthetisierten Strang stößt und sich dann wieder zurückbewegt. Diese Schritte erzeugen kleine DNA-Sequenzfragmente, die als Okazaki-Fragmente bekannt sind und jeweils durch einen RNA-Primer getrennt sind. Der Strang mit den Okazaki-Fragmenten wird als nacheilender Strang bezeichnet und seine Synthese soll diskontinuierlich erfolgen. Nach der Synthese durch DNA-Polymerase III werden die Primer durch die Exonukleaseaktivität von DNA-Polymerase I entfernt und die Lücken gefüllt. Die Lücken, die zwischen der neu synthetisierten DNA (die den RNA-Primer ersetzt hat) und der zuvor synthetisierten DNA verbleiben, sind versiegelt durch das Enzym DNA-Ligase, das die Bildung einer kovalenten Phosphodiesterbindung zwischen dem 3'-OH-Ende eines DNA-Fragments und dem 5'-Phosphatende des anderen Fragments katalysiert und so das Zucker-Phosphat-Rückgrat des DNA-Moleküls stabilisiert. Replikation in Eukaryoten Eukaryontische Genome sind viel komplexer und größer als prokaryontische Genome und bestehen typischerweise aus mehreren linearen Chromosomen. Das menschliche Genom verfügt beispielsweise über 3 Milliarden Basenpaare pro haploidem Chromosomensatz, und 6 Milliarden Basenpaare werden während der Replikation eingefügt. Auf jedem eukaryotischen Chromosom gibt es mehrere Replikationsursprünge. Das menschliche Genom hat 30.000 bis 50.000 Replikationsursprünge. Die Replikationsrate beträgt etwa 100 Nukleotide pro Sekunde – zehnmal langsamer als die prokaryotische Replikation. Die wesentlichen Schritte der Replikation sind bei Eukaryoten die gleichen wie bei Prokaryoten. Bevor die Replikation beginnen kann, muss die DNA als Vorlage zur Verfügung gestellt werden. Eukaryotische DNA ist hochgradig superspiralisiert und verpackt, was durch viele Proteine, einschließlich Histone, erleichtert wird. Nach dem Beginn der Replikation wird die Verlängerung durch eukaryotische DNA-Polymerasen in einem ähnlichen Prozess wie bei Prokaryoten erleichtert. Der führende Strang wird kontinuierlich durch das eukaryontische Polymeraseenzym pol δ synthetisiert, während der nacheilende Strang durch pol ε synthetisiert wird. Das Enzym Ribonuklease H (RNase H) entfernt anstelle einer DNA-Polymerase wie bei Bakterien den RNA-Primer, der dann durch DNA-Nukleotide ersetzt wird. Die verbleibenden Lücken werden durch DNA-Ligase verschlossen. Wie bei Prokaryoten kann die eukaryotische DNA-Polymerase Nukleotide nur in der 5’- bis 3’-Richtung hinzufügen. Im Leitstrang wird die Synthese fortgesetzt, bis sie entweder das Ende des Chromosoms oder eine andere Replikationsgabel erreicht, die in die entgegengesetzte Richtung verläuft. Auf dem Folgestrang wird die DNA in kurzen Abschnitten synthetisiert, von denen jeder durch einen separaten Primer initiiert wird. Wenn die Replikationsgabel das Ende des linearen Chromosoms erreicht, gibt es keinen Platz mehr, um einen Primer für das zu kopierende DNA-Fragment am Ende des Chromosoms herzustellen. Diese Enden bleiben daher ungepaart und können mit der Zeit durch die weitere Zellteilung immer kürzer werden. DNA replication copies genetic material before a cell divides. Each original DNA strand serves as a template for a new complementary strand. The result is semiconservative replication, which means each new DNA molecule contains one old strand and one new strand. The two daughter cells then receive DNA with the same sequence.

Replication starts when helicase separates the two strands of the double helix. This creates a Y-shaped replication fork. Primase then adds short RNA primers that give DNA polymerase a place to begin DNA synthesis. In bacteria, the main DNA polymerases are DNA pol I, DNA pol II, and DNA pol III. DNA pol III carries out DNA synthesis, while DNA pol I and DNA pol II mainly help with repair.

DNA pol III adds deoxyribonucleotides one at a time to the 3'-OH end of the growing strand. It can extend DNA only in the 5' to 3' direction. The energy for this process comes from the three phosphate groups on each nucleotide. When a phosphate bond is broken and diphosphate is released, that energy helps form the phosphodiester bond in the DNA backbone.

Because the DNA double helix is antiparallel, the two template strands face opposite directions. The strand copied toward the replication fork is made continuously and is called the leading strand. The other strand is copied away from the fork. It must start over with new primers as the fork opens, so it is made in short stretches called Okazaki fragments. This strand is the lagging strand, and its synthesis is discontinuous.

After DNA pol III finishes, DNA pol I removes the RNA primers in bacteria, and the gaps are filled in. DNA ligase then seals the remaining nicks by forming covalent phosphodiester bonds between adjacent DNA fragments. This step strengthens the sugar-phosphate backbone of the new DNA molecule.

Eukaryotic DNA replication follows the same basic steps, but the genome is larger and more complex. Eukaryotic chromosomes are linear and have many origins of replication. In humans, the genome has 3 billion base pairs per haploid set, and about 6 billion base pairs are copied during replication. Human chromosomes also have about 30,000 to 50,000 origins of replication, and replication occurs at about 100 nucleotides per second, which is much slower than in prokaryotes.

Before replication begins, eukaryotic DNA must be made available as a template. It is tightly packed and supercoiled, and histones help organize this packaging. After initiation, eukaryotic DNA polymerases carry out elongation. Polymerase delta (pol δ) synthesizes the leading strand, and polymerase epsilon (pol ε) synthesizes the lagging strand. RNase H removes the RNA primers, DNA nucleotides replace them, and DNA ligase seals the final gaps. Because eukaryotic DNA polymerase also works only in the 5' to 3' direction, the lagging strand is still made in short pieces. Near the ends of linear chromosomes, there is no place to add a final primer, so these ends remain unpaired and can become shorter over time as cells keep dividing.
