Introduction to Enzymes

Enzyme sind biologische Katalysatoren, die die Reaktionsgeschwindigkeit beschleunigen, ohne verbraucht zu werden. Sie sind typischerweise Proteine, die sich im Zytoplasma, in Organellen und in Zell- oder Organellenmembranen befinden.
Enzyme können intrazellulär funktionieren oder extrazellulär sezerniert werden. Die meisten Enzyme benötigen eine bestimmte Temperatur und einen bestimmten pH-Wert, um mit maximaler Effizienz zu arbeiten.
Substrate sind die Reaktanten, die spezifisch an aktive Zentren binden, kleine Regionen auf dem Enzym, in denen die Reaktion stattfindet. Die restlichen Teile des Enzyms geben dem Enzym Struktur oder interagieren mit anderen Molekülen, um die Reaktion zu fördern oder zu hemmen.
Enzyme verändern ihre Form, um ihre Substrate zu binden und die Umwandlung von Substraten in Produkte zu katalysieren. Nach der Reaktion gibt das Enzym das Produkt frei und kehrt in seine ursprüngliche Konformation zurück, was es dem Enzym ermöglicht, weitere Reaktionsrunden zu katalysieren.
Einige Enzyme funktionieren nur, wenn sie mit Cofaktoren assoziiert sind, die eine enzymkatalysierte Reaktion unterstützen. Nicht-Proteinmoleküle wie Vitamine, Metallionen oder ATP fungieren als Cofaktoren für verschiedene Enzyme.

einführung in enzyme

Die Verwendung von Enzymen durch den Menschen geht auf das Jahr 7000 v. Chr. zurück. Der Mensch nutzte zunächst Enzyme, um Zucker zu fermentieren und Alkohol herzustellen, ohne zu wissen, dass es sich hierbei um eine enzymkatalysierte Reaktion handelte. Wilhelm Kuhne prägte 1877 den Begriff „Enzym“ aus den griechischen Wörtern „en“, was „in“ oder „innerhalb“ bedeutet, und „zyme“, was „Hefe“ bedeutet.

Die meisten Enzyme sind Proteine, die biochemische Reaktionen beschleunigen, ohne verbraucht zu werden. Enzyme enthalten eine oder mehrere aktive Stellen, die die Substrate binden und in Produkte umwandeln. Viele Enzyme haben auch allosterische Stellen, zusätzliche Stellen, an denen andere Moleküle als Substrate binden, um das Enzym zu aktivieren oder zu hemmen.

Einige Enzyme benötigen zur Katalyse die Anwesenheit von Cofaktoren – Metallionen oder organischen Molekülen. Cofaktoren binden an das aktive Zentrum eines Enzyms und unterstützen die Umwandlung von Substraten in Produkte. Ein Enzym ohne Cofaktor ist ein Apoenzym und wird zu einem Holoenzym, sobald es einen Cofaktor bindet. Ein Cofaktor, der ein organisches Molekül ist, wird Coenzym genannt. Die meisten Coenzyme stammen aus Vitaminen.

Inhibitoren sind Moleküle, die eine enzymkatalysierte Reaktion stoppen. Kompetitive Inhibitoren ähneln Substratmolekülen und konkurrieren mit ihnen um die Bindung an die aktiven Zentren. Im Gegensatz dazu binden nichtkompetitive Inhibitoren an allosterische Stellen und ändern die Enzymkonformation, um die Substratbindung zu verringern.

Enzymes are proteins that speed up biochemical reactions without being used up. Humans have used enzymes since about 7000 BCE, when they fermented sugars to make alcohol without knowing the reaction was enzyme-catalyzed. The word enzyme was coined by Wilhelm Kuhne in 1877. It comes from the Greek words en, meaning “in” or “within,” and zyme, meaning “yeast.”

Enzymes work through active sites, which are regions that bind substrates and help convert them into products. Many enzymes also have allosteric sites. These are extra binding sites where molecules other than substrates can attach and either activate or inhibit the enzyme.

Some enzymes need cofactors to carry out catalysis. Cofactors are metal ions or organic molecules that bind to the active site and help the reaction happen. An enzyme without its cofactor is called an apoenzyme. When the cofactor is attached, it becomes a holoenzyme. An organic cofactor is called a coenzyme, and most coenzymes are derived from vitamins.

Enzyme activity can also be blocked by inhibitors. Competitive inhibitors are similar to the substrate and compete for the active site. Non-competitive inhibitors bind to allosteric sites instead. This changes the enzyme’s shape and reduces substrate binding.
