Enzymes and Activation Energy

In a chemical reaction, the activation energy, or Ea, is the difference in free energy between the substrates in their ground state and in their high energy transition state, an unstable transient structure.
These reactants must have sufficient kinetic energy to collide frequently and undergo chemical modifications in order to form products.
Due to the activation energy for a reaction, the transition state has a higher energy than either the reactants or the products. It can quickly lose that energy and return to the reactants or convert into the products.
When substrates bind to their enzymes, the activation energy decreases, allowing more reactants to reach the transition state. This allows more molecules to be transformed into products and increases the rate of an enzyme-catalyzed reaction compared to an uncatalyzed one.
Enzymes speed up reaction rates in several ways, including stabilizing the transition state by positioning the substrates in the proper orientation or providing appropriate chemical environments, such as charge or pH.

The activation energy (or free energy of activation), abbreviated as Ea, is the small amount of energy input necessary for all chemical reactions to occur. During chemical reactions, certain chemical bonds break, and new ones form. For example, when a glucose molecule breaks down, bonds between the molecule&#39;s carbon atoms break. Since these are energy-storing bonds, they release energy when broken. However, the molecule must be somewhat contorted to get into a state that allows the bonds to break. A small energy input is required to achieve this high-energy, unstable state, called the transition state. For this reason, reactant molecules do not last long in their transition state but very quickly proceed to the chemical reaction&#39;s next steps. The reaction&#39;s transition state exists at a higher energy state than the reactants, and thus, Ea is always positive.
When an enzyme binds its substrate, it forms an enzyme-substrate complex. This complex lowers the reaction&#39;s activation energy and promotes its rapid progression in one of many ways. The enzyme-substrate complex can lower the activation energy by contorting substrate molecules in such a way as to facilitate bond-breaking, helping to reach the transition state. Enzymes can also lower activation energies by taking part in the chemical reaction. The amino acid residues can provide specific ions or chemical groups that form covalent bonds with substrate molecules as a necessary step in the reaction process. It is important to remember that the enzyme will always return to its original state at the reaction&#39;s completion. One of the hallmark properties of enzymes is that they remain ultimately unchanged by the reactions they catalyze. After an enzyme catalyzes a reaction, it releases its product(s).

Enzymes lower the energy needed for a chemical reaction to begin. That starting energy is called activation energy, or free energy of activation, and it is often written as Ea. During a reaction, bonds break and new bonds form. In glucose breakdown, for example, bonds between carbon atoms break and release energy because they are energy-storing bonds.

Before a reaction can move forward, reactant molecules must reach a high-energy, unstable shape called the transition state. To get there, the molecules may need to become somewhat contorted. This small energy input is required because the transition state sits at a higher energy level than the reactants. For that reason, activation energy is always positive, and reactants do not stay in the transition state for long.

Enzymes work by binding their substrates and forming an enzyme-substrate complex. This complex helps lower activation energy and speeds up the reaction. In some cases, the enzyme shapes the substrate so bond-breaking becomes easier and the transition state is reached more quickly. In other cases, amino acid residues in the enzyme take part in the reaction by providing specific ions or chemical groups that form covalent bonds with the substrate.

After the reaction finishes, the enzyme releases its product or products. The enzyme returns to its original state and is not permanently changed by the reaction it catalyzes. That unchanged recycling is one of the key features that makes enzymes effective catalysts.
