Catalytically Perfect Enzymes

Catalytically perfect enzymes have the maximum catalytic efficiency with high turnover numbers, kcat, and strong affinities for their substrates, KM. 
&#160;
The kcat to KM ratio measures catalytic efficiency. For catalytically perfect enzymes, this value ranges from 108 to 109 per molar substrate per second. 
&#160;
Substrate diffusion is the constant movement of substrate molecules caused by molecular motion. Substrates must diffuse within the cell to interact with their enzymes. 
&#160;
The reaction rates of catalytically perfect enzymes are limited only by the speed of substrate diffusion into or product release from the active site. Some enzymes have achieved catalytic perfection through electrostatic forces that attract and guide substrates into their active site.

In catalytically perfect enzymes, every enzyme-substrate collision yields an intermediate that quickly transforms into products.&#160;

The theory of catalytically perfect enzymes was first proposed by W.J. Albery and J. R. Knowles in 1976. These enzymes catalyze biochemical reactions at high-speed. Their catalytic efficiency values range from 108-109 M-1s-1. These enzymes are also called &#39;diffusion-controlled&#39;&#160;as the only rate-limiting step in the catalysis is that of the substrate diffusion into the active site. Examples include triose phosphate isomerase, fumarase, and superoxide dismutase.

Most enzymes achieve catalytic perfection due to the charged groups present on their surface that orient and steer the substrate into the active site. Some other enzymes have a specific active site arrangement, contributing to catalytic perfection. In enzymes such as superoxide dismutase, metal ions, such as copper and zinc, in the active site and charged amino acids, such as arginine, close to the active site speed up the conversion of superoxide anion into oxygen and hydrogen peroxide. Random mutations in enzymes favor such interactions with their substrates, and enzymes with higher efficiency are naturally selected over time.&#160;&#160;
Perfect enzymes not only catalyze highly efficient reactions but can also help protect cells from harmful reaction intermediates. For example, triose phosphate isomerase (TPI) is an enzyme in the glycolytic pathway that catalyzes the interconversion of dihydroxyacetone phosphate (DHAP) and glyceraldehyde 3-phosphate (G3P). The slow conversion of DHAP into G3P forms an enediol intermediate that eventually decomposes into a toxic compound. As TPI is a catalytically perfect enzyme,&#160; it speeds up the reaction and quickly transforms the intermediate into the product, avoiding undesirable compounds. So far, very few enzymes have evolved to be catalytically perfect. Most enzymes are moderately efficient.

Catalytically perfect enzymes are enzymes that speed up biochemical reactions at very high rates. The idea was first proposed by W. J. Albery and J. R. Knowles in 1976. These enzymes have catalytic efficiency values of about 10^8 to 10^9 M^-1 s^-1 and are also called diffusion-controlled enzymes because diffusion of the substrate into the active site is the only rate-limiting step.

Examples of catalytically perfect enzymes include triose phosphate isomerase, fumarase, and superoxide dismutase. In many cases, charged groups on the enzyme surface help orient and steer the substrate into the active site. Some enzymes also depend on a specific active site arrangement, which supports this very fast catalysis.

Superoxide dismutase shows how the active site can increase reaction speed. In this enzyme, metal ions such as copper and zinc work with charged amino acids like arginine near the active site. Together, they speed the conversion of superoxide anion into oxygen and hydrogen peroxide.

Catalytic perfection can also arise through natural selection. Random mutations may favor enzyme-substrate interactions, and enzymes with higher efficiency can be selected over time. Even so, only a few enzymes have evolved to this level, and most enzymes are only moderately efficient.

These enzymes can also protect cells from harmful reaction intermediates. Triose phosphate isomerase, or TPI, works in the glycolytic pathway and converts dihydroxyacetone phosphate, or DHAP, into glyceraldehyde 3-phosphate, or G3P. If DHAP changes too slowly, it forms an enediol intermediate that can break down into a toxic compound. Because TPI is catalytically perfect, it rapidly drives the reaction forward and helps prevent unwanted products.
