Enzymes and Activation Energy

Bei einer chemischen Reaktion ist die Aktivierungsenergie oder Ea die Differenz der freien Energie zwischen den Substraten in ihrem Grundzustand und in ihrem Übergangszustand mit hoher Energie, einer instabilen transienten Struktur.
Diese Reaktanten müssen über eine ausreichende kinetische Energie verfügen, um häufig zu kollidieren und chemisch modifiziert zu werden, um Produkte zu bilden.
Aufgrund der Aktivierungsenergie für eine Reaktion hat der Übergangszustand eine höhere Energie als entweder die Reaktanten oder die Produkte. Es kann diese Energie schnell verlieren und zu den Reaktanten zurückkehren oder sich in die Produkte umwandeln.
Wenn Substrate an ihre Enzyme binden, nimmt die Aktivierungsenergie ab, so dass mehr Reaktanten den Übergangszustand erreichen können. Dadurch können mehr Moleküle in Produkte umgewandelt werden und die Geschwindigkeit einer enzymkatalysierten Reaktion im Vergleich zu einer unkatalysierten Reaktion erhöht werden.

Bei chemischen Reaktionen brechen bestimmte chemische Bindungen auf und es entstehen neue. Wenn beispielsweise ein Glukosemolekül zerfällt, brechen die Bindungen zwischen den Kohlenstoffatomen des Moleküls auf. Da es sich um energiespeichernde Bindungen handelt, geben sie beim Aufbrechen Energie frei. Allerdings muss das Molekül etwas verdreht werden, um in einen Zustand zu gelangen, der das Aufbrechen der Bindungen ermöglicht. Um diesen energiereichen, instabilen Zustand, den sogenannten Übergangszustand, zu erreichen, ist ein geringer Energieaufwand erforderlich. Aus diesem Grund verweilen die Reaktantenmoleküle nicht lange in ihrem Übergangszustand, sondern gehen sehr schnell zu den nächsten Schritten der chemischen Reaktion über. Der Übergangszustand der Reaktion liegt in einem höheren Energiezustand als die Reaktanten, und daher ist Ea immer positiv.

Wenn ein Enzym sein Substrat bindet, bildet es einen Enzym-Substrat-Komplex. Dieser Komplex senkt die Aktivierungsenergie der Reaktion und fördert auf viele Arten ihr schnelles Fortschreiten. Der Enzym-Substrat-Komplex kann die Aktivierungsenergie senken, indem er Substratmoleküle so verzerrt, dass das Aufbrechen von Bindungen erleichtert und so das Erreichen des Übergangszustands erleichtert wird. Enzyme können auch die Aktivierungsenergie senken, indem sie an der chemischen Reaktion teilnehmen. Die Aminosäurereste können spezifische Ionen oder chemische Gruppen bereitstellen, die als notwendiger Schritt im Reaktionsprozess kovalente Bindungen mit Substratmolekülen eingehen. how enzymes lower reaction energy

Enzymes lower the energy needed for a chemical reaction to begin. That starting energy is called activation energy, or free energy of activation, and it is often written as Ea. During a reaction, bonds break and new bonds form. In glucose breakdown, for example, bonds between carbon atoms break and release energy because they are energy-storing bonds.

Before a reaction can move forward, reactant molecules must reach a high-energy, unstable shape called the transition state. To get there, the molecules may need to become somewhat contorted. This small energy input is required because the transition state sits at a higher energy level than the reactants. For that reason, activation energy is always positive, and reactants do not stay in the transition state for long.

Enzymes work by binding their substrates and forming an enzyme-substrate complex. This complex helps lower activation energy and speeds up the reaction. In some cases, the enzyme shapes the substrate so bond-breaking becomes easier and the transition state is reached more quickly. In other cases, amino acid residues in the enzyme take part in the reaction by providing specific ions or chemical groups that form covalent bonds with the substrate.

After the reaction finishes, the enzyme releases its product or products. The enzyme returns to its original state and is not permanently changed by the reaction it catalyzes. That unchanged recycling is one of the key features that makes enzymes effective catalysts.
