ATP Energy Storage and Release

Adenosintriphosphat oder ATP ist die wichtigste Energiewährung, die mehrere biochemische Prozesse in einer lebenden Zelle antreibt.
ATP ist eine organische Verbindung, die aus einem Adenosinmolekül besteht, das als A dargestellt wird und an drei Phosphatgruppen gebunden ist, die mit dem Buchstaben P dargestellt sind. Die drei Phosphate sind durch zwei hochenergetische Phosphoanhydrid-Bindungen miteinander verbunden. Die Hydrolyse dieser Bindungen kann je nach den intrazellulären Bedingungen etwa 46 bis 54 Kilojoule pro Mol freier Energie liefern.
Da die Bindung einer Phosphatgruppe an ein ADP-Molekül energetisch ungünstig ist, beziehen die Zellen Energie aus der Photosynthese oder der Zellatmung, um die Phosphoanhydridbindung zwischen ADP und der dritten Phosphatgruppe zu bilden.
Umgekehrt wird ATP bei Energiebedarf zu anorganischem Phosphat und einem ADP-Molekül hydrolysiert. Diese energetisch günstige Reaktion ist an andere ungünstige Reaktionen gekoppelt, bei denen das freigesetzte Phosphat auf den Reaktanten übertragen wird, um ein neues Produkt zu bilden.
Darüber hinaus treibt die durch die ATP-Hydrolyse freigesetzte Energie die Pumpen an, die gelöste Stoffe durch die Membranen bewegen, und treibt auch die Muskelkontraktion und die neuronalen Signalübertragungswege an.

ATP ist ein äußerst instabiles Molekül. Sofern ATP nicht schnell zur Verrichtung von Arbeit genutzt wird, zerfällt es spontan in ADP und anorganisches Phosphat (Pi), und die bei diesem Prozess freigesetzte freie Energie geht als Wärme verloren. Die durch die ATP-Hydrolyse freigesetzte Energie wird zur Verrichtung von Arbeit innerhalb der Zelle verwendet und hängt von einer Strategie namens Energiekopplung ab. Zellen koppeln die exergonische Reaktion der ATP-Hydrolyse mit endergonischen Reaktionen und ermöglichen so deren Ablauf.

Ein Beispiel für die Energiekopplung mithilfe von ATP ist eine Transmembran-Ionenpumpe, die für die Zellfunktion äußerst wichtig ist. Diese Natrium-Kalium-Pumpe (Na+/K+-Pumpe) treibt Natrium aus der Zelle und Kalium in die Zelle. Ein großer Prozentsatz des ATP einer Zelle wird für den Betrieb dieser Pumpe aufgewendet, da zelluläre Prozesse regelmäßig große Mengen Natrium in die Zelle importieren und große Mengen Kalium aus der Zelle exportieren. Die Pumpe arbeitet ständig daran, die zellulären Konzentrationen von Natrium und Kalium zu stabilisieren. Damit die Pumpe einen Zyklus durchführt (drei Na+-Ionen exportiert und zwei K+-Ionen importiert), muss ein ATP-Molekül hydrolysiert werden. Bei der Hydrolyse von ATP wird sein Gammaphosphat auf das Pumpprotein übertragen.

Dieser Vorgang der Bindung einer Phosphatgruppe an ein Molekül wird als Phosphorylierung bezeichnet. Wie bei den meisten Fällen der ATP-Hydrolyse wird ein Phosphat von ATP auf ein anderes Molekül übertragen. Im phosphorylierten Zustand verfügt die Na^+/K^+-Pumpe über mehr freie Energie und wird zu einer Konformationsänderung angeregt. Durch diese Veränderung kann Na^+ nach außen aus der Zelle abgegeben werden. Anschließend bindet es extrazelluläres K^+, was durch eine weitere Konformationsänderung dazu führt, dass sich das Phosphat von der Pumpe löst. Diese Freisetzung von Phosphat löst die Freisetzung von K+ in das Zellinnere aus. Im Wesentlichen ist die bei der Hydrolyse von ATP freigesetzte Energie mit der Energie gekoppelt, die zum Antreiben der Pumpe und zum Transport von Na^+- und K+-Ionen erforderlich ist. ATP führt zelluläre Arbeit aus und nutzt diese grundlegende Form der Energiekopplung durch Phosphorylierung.

Bei zellulären Stoffwechselreaktionen wie der Nährstoffsynthese und dem Nährstoffabbau müssen sich bestimmte Moleküle häufig leicht in ihrer Konformation ändern, um als Substrat für den nächsten Schritt in der Reaktionsreihe zu dienen. Ein Beispiel ist die Glykolyse, die allerersten Schritte der Zellatmung. In diesem ersten Schritt wird ATP benötigt, um Glukose zu phosphorylieren, wodurch ein energiereiches, aber instabiles Zwischenprodukt entsteht. Diese Phosphorylierungsreaktion löst eine Konformationsänderung aus, die es dem phosphorylierten Glucosemolekül ermöglicht, sich in den phosphorylierten Zucker Fructose umzuwandeln. Fruktose ist ein notwendiges Zwischenprodukt, damit die Glykolyse voranschreiten kann. Hier stellt die exergonische Reaktion der ATP-Hydrolyse in Verbindung mit der endergonischen Reaktion der Glucosephosphorylierung einen Zwischenschritt auf dem Weg dar. Wieder einmal wurde die durch das Aufbrechen einer Phosphatbindung innerhalb von ATP freigesetzte Energie zur Phosphorylierung eines anderen Moleküls genutzt, wodurch ein instabiles Zwischenprodukt entstand und eine wichtige Konformationsänderung ausgelöst wurde.

ATP powers cellular transport and many reactions by transferring energy through phosphorylation. ATP is a highly unstable molecule. If it is not used quickly, it breaks down into ADP and inorganic phosphate, and the released energy is lost as heat.

Cells use ATP through energy coupling. This means the exergonic reaction of ATP hydrolysis is linked to an endergonic reaction so the reaction can move forward. One major example is the sodium-potassium pump, a transmembrane ion pump that is vital for cell function.

The sodium-potassium pump moves sodium out of the cell and potassium into the cell. Cells spend a large amount of ATP on this pump because sodium and potassium are regularly moved in opposite directions across the membrane. To complete one cycle, the pump exports three sodium ions and imports two potassium ions, and one ATP molecule must be hydrolyzed.

During ATP hydrolysis, the gamma phosphate is transferred to the pump protein. This is called phosphorylation, which means a phosphate group binds to a molecule. In the phosphorylated state, the pump has more free energy and changes shape. That shape change lets it release sodium outside the cell. The pump then binds potassium outside the cell, and another shape change removes the phosphate and releases potassium inside the cell.

ATP also helps start steps in cellular metabolism. In glycolysis, which is the first stage of cellular respiration, ATP phosphorylates glucose. This creates a high-energy but unstable intermediate. The phosphorylation also drives a conformational change that helps the molecule become phosphorylated fructose, which is needed for glycolysis to continue. In both examples, ATP hydrolysis is coupled to another reaction by transferring a phosphate group and powering an important change in shape.
