Die Cope-Eliminierung, ähnlich wie die Hofmann-Eliminierung, erzeugt das weniger substituierte Alken als Hauptprodukt, indem N,N-Dimethylhydroxylamin aus einem Aminoxid anstelle eines tertiären Amins aus einem quartären Ammoniumsalz eliminiert wird.
Das Aminoxid entsteht aus der Oxidation eines tertiären Amins durch Wasserstoffperoxid.
Die volle positive Ladung am Stickstoffatom fördert die Cope-Eliminierung, bei der es sich um einen konzertierten Eliminierungsprozess handelt, der der Synstereochemie unter thermischen Bedingungen folgt.
Das Aminoxid abstrahiert intramolekular das weniger behinderte β Proton, gefolgt von einem zyklischen Elektronenfluss, der schließlich zum Verlust der Abgangsgruppe und zur Bildung des weniger substituierten Alkens führt.
Die am Übergangszustand beteiligten Atome sind nahezu planar angeordnet, wobei das β Wasserstoff und die Verlassensgruppe syn orientiert sind.
Die Cope-Eliminierung erfordert im Gegensatz zur Hofmann-Eliminierung keine externe Base, da das Aminoxid als Base in der Reaktion fungiert.

Der Mechanismus der Reaktion ist in Abbildung 2 dargestellt. Hier ergibt die Oxidation tertiärer Amine durch Wasserstoffperoxid Aminoxid. Anschließend erfolgt die thermisch induzierte Abspaltung des N,N-Dimethylhydroxylamins als Abgangsgruppe. Als Hauptprodukt entsteht jedoch ein weniger substituiertes Alken. In diesem Prozess folgt Aminoxid in der Rolle einer Base der syn-Stereochemie und durchläuft einen zyklischen Übergangszustand.

The Cope elimination reaction converts tertiary amines into alkenes. It uses hydrogen peroxide and heat. The process starts with oxidation of the tertiary amine to form an amine oxide.

The amine oxide then acts as a base during the elimination step. The reaction follows syn stereochemistry, which means the groups leave from the same side. It also proceeds through a cyclic transition state. The N,N-dimethyl hydroxylamine group is formed as the leaving group.

The major product is usually the less substituted alkene. Cope elimination is useful when reactive or sensitive alkenes are needed. It is valued because it takes place under mild conditions.
