Arenediazonium salts, when treated with substituted aromatic rings bearing activating groups, undergo a coupling reaction to generate compounds bearing an &#8211;N=N&#8211; group.
This is an electrophilic aromatic substitution reaction, wherein the electrophilic diazonium ion reacts at the para position of the nucleophilic aromatic compound, resulting in the resonance-stabilized carbocation.
Next, deprotonation of the carbocation leads to the formation of the coupled product.
If the para position of the aromatic ring is unavailable, coupling occurs at the ortho position.
Colored azo compounds are often used as synthetic dyes and pH indicators. For example, &#39;sunset yellow&#39; is a dye responsible for the yellow color of ice cream.
Methyl orange is used as an acid&#8211;base indicator during titrimetric analysis. In the presence of an alkaline solution, it turns into a yellow-colored solution, while in acidic media, upon protonation, the color changes to red.

The reaction of weakly electrophilic aryldiazonium (also called arenediazonium) salts with highly activated aromatic compounds leads to the formation of products with an &#8212;N=N&#8212; link, called an azo linkage. This reaction, presented in Figure 1, is known as diazo coupling and occurs without the loss of the nitrogen atoms of the aryldiazonium salt. Highly activated aromatic compounds such as phenols or arylamines favor the diazo coupling reaction. The coupling generally occurs at the para position. However, if the para position is substituted, the coupling occurs instead at the ortho position.

Mechanistically, the reaction proceeds via an electrophilic substitution reaction via donation of the &#960; electrons of the activated aromatic nucleus to the electrophilic nitrogen, resulting in a resonance-stabilized carbocation.

Figure 3 depicts the final step, which involves the deprotonation of the carbocation to yield the diazo product.

As depicted in Figure 4, the role of basicity in diazo coupling reaction is evident. For phenols, it is most feasible under slightly alkaline conditions, where the phenol is present as phenoxide ions. The formation of appreciable phenoxide ions activates the aromatic ring, thereby enhancing the coupling between arenediazonium ions and phenols in a slightly basic solution. However, excess alkalinity of the solution (pH &#62; 10) leads to the formation of diazohydroxide or diazotate ions, which are unreactive towards coupling.

Unlike phenols, as shown in Figure 5, the diazo coupling of amines occurs optimally in the acidic pH range of 5&#8211;7. The reaction is driven by the concentration of the arenediazonium ion, which is at a maximum at this pH.

Diazo-coupled compounds feature an extended conjugated &#960; electron system by aligning the two aromatic rings in conjugation, making them strongly chromophoric. This results in the intense color of the azo compounds known as azo dyes. Examples of azo dyes include alizarin yellow, butter yellow, congo red, orange II, and para red.
Apart from being an excellent dye, diazo compounds were also identified as antibiotics when Domagk G., in 1935, successfully used prontosil on his daughter to prevent lethal streptococcal infection, which marked the beginning of the modern era of synthetic antibiotics. Prontosil is inactive against bacteria, but in the human body, it metabolizes to give a compound called sulfanilamide, which exhibits antibacterial properties.

Diazo coupling is the reaction that forms azo dyes from aryldiazonium salts and highly activated aromatic compounds. The product contains an —N=N— azo linkage, and the nitrogen atoms from the diazonium salt remain in the product. Phenols and arylamines are common activated partners for this reaction.

The coupling usually takes place at the para position on the aromatic ring. If the para position is already occupied, the reaction shifts to the ortho position instead. The mechanism is an electrophilic substitution reaction. The activated aromatic ring donates pi electrons to the electrophilic nitrogen, forms a resonance-stabilized carbocation, and then loses a proton to give the diazo product.

Basicity affects diazo coupling in important ways. For phenols, the reaction works best under slightly alkaline conditions, where phenoxide ions form and activate the ring. If the solution becomes too basic, with pH above 10, diazohydroxide or diazotate ions form and do not couple well.

Amines behave differently. Their diazo coupling is most effective in the acidic pH range of 5 to 7, where the concentration of the arenediazonium ion is highest. The final azo compounds have an extended conjugated pi system because the two aromatic rings are aligned in conjugation. This makes them strongly chromophoric, which gives azo dyes their intense color.

Common examples of azo dyes include alizarin yellow, butter yellow, congo red, orange II, and para red. Diazo compounds are also linked to medicine. In 1935, Domagk used prontosil to help prevent a lethal streptococcal infection in his daughter, which marked the start of the modern era of synthetic antibiotics. Prontosil itself is inactive, but in the body it is converted to sulfanilamide, the antibacterial form.
