Sekundäre Amine – einschließlich Alkyl und Aryl – ergeben nach Behandlung mit NaNO2 und einer starken Säure sekundäre N-Nitrosamine.
Es sei daran erinnert, dass eine angesäuerte Lösung von NaNO2 HNO2 erzeugt. Das instabile HNO2 wird weiter protoniert und anschließend dehydriert, um das Nitrosonium-Ion zu bilden – ein Elektrophil.
Das in situ erzeugte Nitrosylkation wird von den nukleophilen sekundären Aminen angegriffen und bildet ein N-Nitrosammonium-Ion.
Das Ion führt bei der Deprotonierung durch Lösungsmittelmoleküle zur Bildung von sekundären N-Nitrosaminen.
Im Gegensatz zu primären N-Nitrosaminen, die von primären Aminen abgeleitet sind, sind sekundäre N-Nitrosamine stabil, da das Fehlen des N-H-Protons weitere Tautomerie und schließlich die Bildung von Diazonium verhindert.
Sie trennen sich in der Regel als ölige Flüssigkeiten aus dem Reaktionsgemisch.
Im Allgemeinen haben sekundäre N-Nitrosamine seltene synthetische Anwendungen. Viele von ihnen sind jedoch als krebserregend bekannt, die in verarbeiteten Lebensmitteln oder Tabak vorkommen.

Sekundäre Amine reagieren mit salpetriger Säure zu N-Nitrosaminen, wie in Abbildung 1 dargestellt. Salpetrige Säure, eine schwache und instabile Säure, wird in situ aus einer wässrigen Lösung von Natriumnitrit und starken Säuren wie Salzsäure oder Schwefelsäure unter kalten Bedingungen gebildet. In Gegenwart einer Säure wird die salpetrige Säure protoniert. Der anschließende Wasserverlust führt zur Bildung des als Nitrosoniumion bekannten Elektrophils.

Das sekundäre Amin, das als Nukleophil fungiert, reagiert mit dem Nitrosoniumion und ergibt ein weiteres Ion, das als N-Nitrosammoniumion bekannt ist. Das resultierende Ion wird dann durch Wasser deprotoniert, um sekundäre N-Nitrosamine zu bilden. Dieser Reaktionsmechanismus ist in Abbildung 2 dargestellt. Die sekundären N-Nitrosamine lassen sich als ölige gelbe Flüssigkeiten aus dem Reaktionsgemisch abtrennen.

Normalerweise sind sekundäre N-Nitrosamine für die kommerzielle Nutzung von geringerer Bedeutung. Sie werden jedoch aufgrund ihrer starken krebserzeugenden Eigenschaften untersucht. In Norwegen (1962) wurde ein Ausbruch einer Lebensmittelvergiftung bei Schafen auf mit Nitrit behandeltes Fischmehl zurückgeführt. Natriumnitrit wird zur Konservierung verschiedener Lebensmittel und Fleischsorten verwendet. Im Magen reagieren sie mit der Magensäure zu salpetriger Säure. Die salpetrige Säure reagiert dann mit sekundären Aminen in der Nahrung zu stark krebserregenden N-Nitrosaminen. Aus diesem Grund wurde die Verwendung von Natriumnitrit bei der Fleischkonservierung von der FDA auf 50 bis 125 ppm begrenzt. N-Nitrosamine kommen auch im Zigaretten- und Tabakrauch vor.

nitrosamine formation from secondary amines

Secondary amines can form N-nitrosamines when they react with nitrous acid. Nitrous acid is weak and unstable, so it is made in the reaction mixture from sodium nitrite and a strong acid such as hydrochloric acid or sulfuric acid. The reaction is carried out in cold conditions.

In acid, nitrous acid becomes protonated. It then loses water and forms the nitrosonium ion, a reactive electrophile. The secondary amine acts as a nucleophile and attacks this ion. That step gives an N-nitrosammonium ion.

Water then removes a proton from the ion to make the secondary N-nitrosamine. These products can be separated from the mixture as oily yellow liquids. Although they are not usually important for commercial use, they are studied because many are potent carcinogens.

The transcript also links this chemistry to food safety. In 1962, a food-poisoning outbreak in sheep in Norway was traced to nitrite-treated fish meal. Sodium nitrite is used to preserve foods and meat, but in the stomach it can form nitrous acid. That acid can react with secondary amines in food to produce highly carcinogenic N-nitrosamines, so FDA limits sodium nitrite in meat preservation to 50 to 125 ppm. N-nitrosamines are also found in cigarette and tobacco smoke.
