Wenn Arendiazoniumsalze mit substituierten aromatischen Ringen behandelt werden, die aktivierende Gruppen tragen, durchlaufen sie eine Kupplungsreaktion, um Verbindungen zu erzeugen, die eine –N=N– Gruppe tragen.
Hierbei handelt es sich um eine elektrophile aromatische Substitutionsreaktion, bei der das elektrophile Diazoniumion an der para-Position der nukleophilen aromatischen Verbindung reagiert, was zu der resonanzstabilisierten Carbokation führt.
Als nächstes führt die Deprotonierung der Carbokation zur Bildung des gekoppelten Produkts.
Wenn die para-Position des aromatischen Rings nicht verfügbar ist, erfolgt die Kopplung an der ortho-Position.
Farbige Azoverbindungen werden häufig als synthetische Farbstoffe und pH-Indikatoren verwendet. Zum Beispiel ist "Sunset Yellow" ein Farbstoff, der für die gelbe Farbe von Eiscreme verantwortlich ist.
Methylorange wird als Säure-Base-Indikator bei der titrimetrischen Analyse verwendet. In Gegenwart einer alkalischen Lösung verwandelt es sich in eine gelb gefärbte Lösung, während sich die Farbe in sauren Medien bei der Protonierung in Rot ändert.

Aryldiazonium Salts to Azo Dyes: Diazo Coupling

Die Reaktion schwach elektrophiler Aryldiazoniumsalze (auch Arendiazoniumsalze genannt) mit hochaktivierten aromatischen Verbindungen führt zur Bildung von Produkten mit einer -N=N-Verknüpfung, einer sogenannten Azoverknüpfung. Diese in Abbildung 1 dargestellte Reaktion wird als Diazokopplung bezeichnet und läuft ohne den Verlust der Stickstoffatome des Aryldiazoniumsalzes ab. Hochaktivierte aromatische Verbindungen wie Phenole oder Arylamine begünstigen die Diazokopplungsreaktion. Die Kopplung erfolgt im Allgemeinen an der Para-Position. Wenn jedoch die Para-Position ersetzt wird, erfolgt die Kopplung stattdessen an der Ortho-Position.

Mechanistisch gesehen verläuft die Reaktion über eine elektrophile Substitutionsreaktion durch Abgabe der π-Elektronen des aktivierten aromatischen Kerns an den elektrophilen Stickstoff, was zu einem resonanzstabilisierten Carbokation führt.

In Abbildung 3 ist der letzte Schritt dargestellt, bei dem das Carbokation deprotoniert wird, um das Diazoprodukt zu erhalten.

Wie in Abbildung 4 dargestellt, ist die Rolle der Basizität bei der Diazokopplungsreaktion offensichtlich. Bei Phenolen ist dies am besten unter leicht alkalischen Bedingungen möglich, bei denen das Phenol in Form von Phenoxidionen vorliegt. Die Bildung nennenswerter Phenoxidionen aktiviert den aromatischen Ring und verstärkt dadurch die Kopplung zwischen Arendiazoniumionen und Phenolen in einer leicht basischen Lösung. Eine übermäßige Alkalität der Lösung (pH &gt; 10) führt jedoch zur Bildung von Diazohydroxid- oder Diazotationen, die gegenüber der Kupplung nicht reaktiv sind.

Im Gegensatz zu Phenolen erfolgt die Diazokopplung von Aminen, wie in Abbildung 5 dargestellt, optimal im sauren pH-Bereich von 5–7. Die Reaktion wird durch die Konzentration des Arendiazoniumions angetrieben, die bei diesem pH-Wert ihr Maximum erreicht.

Diazogekoppelte Verbindungen zeichnen sich durch ein erweitertes konjugiertes π-Elektronensystem aus, indem die beiden aromatischen Ringe in Konjugation ausgerichtet werden, was sie stark chromophor macht. Dadurch entsteht die intensive Farbe der Azoverbindungen, sogenannte Azofarbsubstanzen. Beispiele für Azofarbsubstanzen sind Alizaringelb, Buttergelb, Kongorot, Orange II und Pararot.

Diazoverbindungen sind nicht nur ein hervorragender Farbstoff, sondern wurden auch als Antibiotika identifiziert, als Domagk G. 1935 erfolgreich Prontosil bei seiner Tochter einsetzte, um eine tödliche Streptokokkeninfektion zu verhindern, was den Beginn der modernen Ära der synthetischen Antibiotika markierte. Prontosil ist gegen Bakterien inaktiv, wird aber im menschlichen Körper zu einer Verbindung namens Sulfanilamid verstoffwechselt, die antibakterielle Eigenschaften aufweist.

diazo coupling and azo dye formation

Diazo coupling is the reaction that forms azo dyes from aryldiazonium salts and highly activated aromatic compounds. The product contains an —N=N— azo linkage, and the nitrogen atoms from the diazonium salt remain in the product. Phenols and arylamines are common activated partners for this reaction.

The coupling usually takes place at the para position on the aromatic ring. If the para position is already occupied, the reaction shifts to the ortho position instead. The mechanism is an electrophilic substitution reaction. The activated aromatic ring donates pi electrons to the electrophilic nitrogen, forms a resonance-stabilized carbocation, and then loses a proton to give the diazo product.

Basicity affects diazo coupling in important ways. For phenols, the reaction works best under slightly alkaline conditions, where phenoxide ions form and activate the ring. If the solution becomes too basic, with pH above 10, diazohydroxide or diazotate ions form and do not couple well.

Amines behave differently. Their diazo coupling is most effective in the acidic pH range of 5 to 7, where the concentration of the arenediazonium ion is highest. The final azo compounds have an extended conjugated pi system because the two aromatic rings are aligned in conjugation. This makes them strongly chromophoric, which gives azo dyes their intense color.

Common examples of azo dyes include alizarin yellow, butter yellow, congo red, orange II, and para red. Diazo compounds are also linked to medicine. In 1935, Domagk used prontosil to help prevent a lethal streptococcal infection in his daughter, which marked the start of the modern era of synthetic antibiotics. Prontosil itself is inactive, but in the body it is converted to sulfanilamide, the antibacterial form.
