Amine sind polare Verbindungen mit einem einsamen Paar am Stickstoffatom, das das gesamte molekulare Dipolmoment bestimmt. Sie wirken als Wasserstoffbrückenakzeptoren in hydroxylischen Lösungsmitteln.
Typischerweise sind niedermolekulare Amine – mit weniger als fünf Kohlenstoffatomen – in Wasser mischbar oder wasserlöslich. Im Vergleich dazu sind hochmolekulare Amine entweder sparsam oder völlig unlöslich in Wasser, da die unpolaren Kohlenwasserstoffketten mit polaren Lösungsmitteln unverträglich sind.
Sowohl primäre als auch sekundäre Amine sind Wasserstoffbrückendonatoren und bilden intermolekulare Wasserstoffbrückenbindungen. Wasserstoffbrückenbindungen beeinflussen direkt die beobachteten Trends in den Siedepunkten.
Daher haben primäre Amine – mit zwei N–H-Bindungen – höhere Siedepunkte als isomere sekundäre und tertiäre Amine.
Intermolekulare Wasserstoffbrückenbindungen in Aminen sind vergleichsweise schwächer als in Alkoholen.
Primäre und sekundäre Amine haben also niedrigere Siedepunkte als ihre Alkoholanaloga, aber höhere Siedepunkte als die Verbindungen, die keine Wasserstoffbrückenbindungen eingehen können.
Amine haben charakteristische Gerüche. Einfache Amine haben einen typischen fischähnlichen Geruch, während einige Diamine einen stechenden Geruch haben; Der Geruch beeinflusst die gebräuchlichen Namen solcher Amine.

Amine mit niedrigem Molekulargewicht sind bei Raumtemperatur in der Regel gasförmig, während Amine mit hohem Molekulargewicht in flüssiger oder fester Form vorliegen. Amine mit niedrigem Molekulargewicht haben in der Regel einen fischartigen Geruch. Diamine haben in der Regel einen stechenden Geruch. Cadaverin und Putrescin, die in Abbildung 1 dargestellt sind, sind zwei Moleküle, die für die Verwesung von Gewebe verantwortlich sind.

Amine sind polar. Dementsprechend ermöglichen primäre und sekundäre Amine mit N-H-Bindungen intermolekulare Wasserstoffbrückenbindungs-Wechselwirkungen. Diese Wechselwirkung führt dazu, dass Amine einen höheren Siedepunkt haben als Alkane oder Ether mit vergleichbarem Molekulargewicht, die nur schwächere Wechselwirkungen wie van-der-Waals-Kräfte oder Dipol-Dipol-Wechselwirkungen aufweisen. Andererseits haben Alkohole einen höheren Siedepunkt als Amine, weil Stickstoff weniger elektronegativ ist als Sauerstoff, was zu schwächeren intermolekularen Wasserstoffbrückenbindungen zwischen Stickstoff und Wasserstoff als zwischen Sauerstoff und Wasserstoff führt. Dieses Schema ist in Abbildung 2 dargestellt.

Wie in Abbildung 3 dargestellt, haben tertiäre Amine niedrigere Siedepunkte als primäre oder sekundäre Amine. Im Gegensatz zu primären oder sekundären Aminen ist bei tertiären Aminen durch das Fehlen der N-H-Bindung keine Wasserstoffbrückenbindung möglich.

Amine mit niedrigem Molekulargewicht sind auch in Wasser löslich - die Wasserstoffbrückenbindungen zwischen Aminen und Wasser, wie in Abbildung 4 dargestellt, führen zu dieser Löslichkeit. Die Wasserlöslichkeit von Aminen nimmt mit zunehmendem Molekulargewicht ab. Im Allgemeinen sind Amine mit weniger als fünf Kohlenstoffatomen wasserlöslich. Aufgrund der Wechselwirkung zwischen dem Elektronenpaar des Stickstoffatoms und dem Wasserstoffatom des Wassers sind auch tertiäre Amine in Wasser löslich.

Im Allgemeinen sind Amine sehr giftig und können bei Verschlucken aufgrund ihrer physiologischen Aktivität zu Vergiftungen oder zum Tod führen. Beispielsweise sind Arylamine wie Benzidin und β-Naphthylamin, die in Abbildung 5 dargestellt sind, krebserregend und sollten mit Vorsicht behandelt werden.

Darüber hinaus wurden in den Hautsekreten des südamerikanischen Pfeilgiftfrosches Phyllobates terribilis zwei giftige Aminverbindungen beobachtet: Batrachotoxin und Batrachotoxinin A. Diese Amine sind so giftig, dass bereits eine Menge von 200 µg ausreicht, um beim Menschen einen irreversiblen Herzstillstand zu verursachen.

Amines show clear physical properties that depend on molecular size and structure. Low molecular weight amines are usually gases at room temperature, while higher molecular weight amines are often liquids or solids. Many low molecular weight amines also have a rotten fish-like smell, and diamines usually have a pungent odor.

Amines are polar molecules, so they can take part in intermolecular hydrogen bonding. Primary and secondary amines have N–H bonds that allow these interactions. Because of this, they usually have higher boiling points than alkanes or ethers with similar molecular weight, since those compounds rely on weaker van der Waals forces or dipole-dipole interactions.

Alcohols still boil at higher temperatures than amines. Nitrogen is less electronegative than oxygen, so the hydrogen bonding between nitrogen and hydrogen is weaker than the hydrogen bonding between oxygen and hydrogen. Tertiary amines have lower boiling points than primary or secondary amines because they do not have an N–H bond, so they cannot hydrogen bond in the same way.

Water solubility also changes with size. Low molecular weight amines are soluble in water because they can form hydrogen bonds with water molecules. In general, amines with fewer than five carbon atoms are water-soluble, and even tertiary amines can dissolve because the lone pair on nitrogen can interact with the hydrogen atom of water. As molecular weight increases, solubility decreases.

Amines can also be dangerous. In general, they are significantly toxic and may cause poisoning or death if swallowed because of their physiological activity. Some arylamines, such as benzidine and beta-naphthylamine, are carcinogenic and must be handled carefully. The poison dart frog Phyllobates terribilis also secretes toxic amine compounds, including batrachotoxin and batrachotoxinin A, and as little as 200 micrograms can cause irreversible cardiac arrest in humans.
