Turnover Number and Catalytic Efficiency

Die Umsatzzahl oder kcat gibt an, wie schnell ein Enzym Substratmoleküle in Produkte umwandeln kann.
kcat ist gleich der maximalen Anzahl von Substratmolekülen, die in einer bestimmten Zeit pro aktivem Zentrum des Enzyms transformiert werden können.
Die Umsatzzahlen verschiedener Enzyme reichen von weniger als einem Substratmolekül bis hin zu Millionen von Molekülen pro Sekunde.
Zur Berechnung von kcat wird die maximale Geschwindigkeit oder Vmax einer enzymkatalysierten Reaktion durch die Gesamtenzymkonzentration dividiert.
Sowohl kcat, die katalytische Rate, als auch KM, die Affinität zu einem Substrat, beeinflussen die katalytische Effizienz eines Enzyms – wie effektiv ein Enzym eine bestimmte biochemische Reaktion beschleunigt.
Eine Möglichkeit, die katalytische Effizienz für ein bestimmtes Substrat zu messen, ist das Verhältnis von kcat zu KM.
Enzyme mit einem hohen k-Gehalt katalysieren schnell die Umwandlung eines Substrats, und Enzyme mit einem niedrigen K-M-Gehalt binden ihre Substrate stark. Daher sind die Enzyme mit einem größeren Verhältnis effizienter.

umsatzzahl und katalytische effizienz

Die Umsatzzahl eines Enzyms ist die maximale Anzahl an Substratmolekülen, die es pro Zeiteinheit umwandeln kann. Die Umsatzzahlen der meisten Enzyme liegen zwischen 1 und 1000 Molekülen pro Sekunde. Katalase hat die bekannte höchste Umsatzzahl und ist in der Lage, bis zu 2,8×106 Moleküle Wasserstoffperoxid pro Sekunde in Wasser und Sauerstoff umzuwandeln. Lysozym hat die niedrigste bekannte Umsatzzahl von einem halben Molekül pro Sekunde.

Chymotrypsin ist ein Pankreasenzym, das bei der Verdauung Proteine abbaut. Die Umsatzzahl von Chymotrypsin beträgt 100 Moleküle pro Sekunde. Würde diese Reaktion unkatalysiert ablaufen, würde es Hunderte von Jahren dauern, bis die Peptidbindungen in Wasser bei neutralem pH-Wert aufbrechen. Somit trägt die hohe Umsatzzahl von Chymotrypsin zu einer schnellen Verdauung von Proteinen im Darm bei.

Das Enzym Ribulose-1,5-Bisphosphat-Carboxylase-Oxygenase oder RuBisCO hat eine sehr geringe Umsatzzahl bei der Fixierung von 3 CO2-Molekülen pro Sekunde und ist eines der langsamsten Enzyme. Allerdings gleicht die Häufigkeit von RuBisCO in der Natur die geringe Umsatzzahl aus. RuBisCO macht etwa 50 % des gesamten in Blättern vorkommenden Proteins aus.

Ein Enzym mit einer hohen Umsatzzahl muss nicht unbedingt hocheffizient sein. Die katalytische Effizienz eines Enzyms wird durch das Verhältnis der Umsatzzahl kcat zur Affinität KM angegeben. Mit anderen Worten: Um effizient zu sein, sollte ein Enzym auch einen geringen KM für das Substrat aufweisen. Die durchschnittliche katalytische Effizienz der meisten Enzyme beträgt etwa 105 M-1s-1, was bedeutet, dass sie mäßig effizient sind. Nur wenige Enzyme mit einer katalytischen Effizienz zwischen 108 und 109 M-1s-1 sind supereffizient oder katalytisch perfekt.

how fast enzymes work in digestion

Enzymes can work at very different speeds, and their turnover number shows how fast they convert substrate molecules into products. Turnover number is the maximum number of substrate molecules an enzyme can transform per unit time. For most enzymes, this value is between 1 and 1000 molecules per second.

Some enzymes are especially fast. Catalase has the highest known turnover number and can convert up to 2.8×10^6 molecules of hydrogen peroxide into water and oxygen each second. Lysozyme has the lowest known turnover number, at half a molecule per second.

Digestion also depends on enzyme speed. Chymotrypsin is a pancreatic enzyme that breaks down proteins during digestion, and its turnover number is 100 molecules per second. Without an enzyme, peptide bonds would take hundreds of years to break in water at neutral pH. Chymotrypsin helps make protein digestion in the intestine much faster.

Not every slow enzyme is unimportant. RuBisCO, or ribulose 1,5-bisphosphate carboxylase oxygenase, has a very low turnover number and fixes only 3 molecules of CO2 per second. It is one of the slowest enzymes, but it is very abundant in nature. RuBisCO makes up around 50% of the total protein found in leaves.

A high turnover number does not always mean high catalytic efficiency. Catalytic efficiency is the ratio of turnover number, kcat, to affinity, KM. An enzyme also needs a low KM for its substrate to be efficient. The average catalytic efficiency of most enzymes is about 10^5 M^-1 s^-1, so they are moderately efficient. A few enzymes reach 10^8 to 10^9 M^-1 s^-1 and are called superefficient or catalytically perfect.
