Das Protonen-NMR-Spektrum eines Amins zeigt das N–H-Signal als Breitband im Bereich von 0,5 bis 4 ppm.
Die Position des Peaks hängt vom Ausmaß der Wasserstoffbrückenbindung, der Art des Lösungsmittels, der Konzentration des Amins und den Temperaturbedingungen ab.
Da die N-H-Protonen leicht austauschbar sind, findet keine Kopplung mit den benachbarten Protonen statt – der Peak ist breit, ungespalten und schwer zu identifizieren.
Ein D2O-Shake tauscht die N-H-Protonen gegen die N-D-Deuteronen aus, wodurch der N-H-Peak verschwindet, was das Vorhandensein von Aminprotonen in der Probe bestätigt.
Da die α Kohlenstoffprotonen durch Stickstoff abgeschirmt werden, haben sie höhere chemische Verschiebungswerte als die β Kohlenstoffprotonen, die weit weniger abgeschirmt sind und weiter oben erscheinen.
Im 13-Grad-NMR-Spektrum zeigt der α Kohlenstoff die höchste chemische Verschiebung, da er durch das Stickstoffatom stark abgeschirmt wird.
Ein zunehmender Abstand zwischen Kohlenstoff und Stickstoff verringert den Entschirmungseffekt, wodurch der chemische Verschiebungswert des Kohlenstoffatoms abnimmt.

Da Aminprotonen in Lösung einen schnellen Protonenaustausch durchlaufen, sind die Protonen labil und nehmen daher an keiner Spaltung mit benachbarten Protonen teil. Daher ist der beobachtete Peak breit und liefert keine Informationen über die angrenzende Protonenumgebung. Dieses Problem wird gelöst und N-H-Protonen werden identifizierbar, indem der Mischung D_2O hinzugefügt wird. Die Zugabe führt dazu, dass die N-H-Protonen mit Deuteronen ausgetauscht werden, was zum Verschwinden der N-H-Protonenpeaks führt. Dieses Verschwinden weist auf das Vorhandensein labiler Protonen in der Probe hin.

In aliphatischen Aminen werden die α-Protonen durch das elektronenziehende Stickstoffatom entschirmt. Folglich zeigen die α-Protonen höhere chemische Verschiebungen (δ 2,2 bis 2,9 ppm) als β-Protonen (δ 1 bis 1,7 ppm), die aufgrund des größeren Abstands vom Stickstoffatom weniger entschirmt sind.

In der ^13C-NMR-Spektroskopie zeigen die α-Kohlensubstanzen aliphatischer Amine aufgrund der entschirmenden Wirkung von elektronegativem Stickstoff die höchsten chemischen Verschiebungswerte im Bereich von δ 30 bis 60 ppm.

Amines in NMR spectroscopy show clear signal patterns that help identify their structure. In proton NMR, primary amines and secondary amines give a broad N–H signal between δ 0.5 and 5 ppm. The exact position can change with sample concentration, hydrogen bonding, and the solvent used.

The N–H protons in amines are labile, which means they exchange quickly in solution. Because of this fast proton exchange, they do not split with nearby protons. Their peak stays broad and does not reveal the environment of adjacent protons.

A D2O test can make these N–H protons easier to identify. When deuterium oxide is added, the N–H protons exchange with deuterons and the N–H peaks disappear. This loss of signal shows that labile protons are present in the sample.

Aliphatic amines also shift nearby carbon and hydrogen signals. The α protons, which are next to the nitrogen atom, are deshielded and appear at higher chemical shifts, about δ 2.2 to 2.9 ppm. The β protons are farther away and appear at lower shifts, about δ 1 to 1.7 ppm. In 13C NMR spectroscopy, the α carbons of aliphatic amines give the highest chemical shift values, from δ 30 to 60 ppm, because nitrogen pulls electron density away.
