Bei Komplexierungsreaktionen bilden Metallkationen Komplexe mit Verbindungen, die als Liganden bezeichnet werden, mit ungeteilten Elektronenpaaren, die die Koordinationszahl der Metallionen erfüllen.
Das Metallkation ist eine Lewis-Säure, ein Elektronenpaarakzeptor, während der Ligand eine Lewis-Base, ein Elektronenpaardonor, ist und die zwischen ihnen gebildete Bindung der Dativ oder die kovalente Koordinierungsbindung ist. 
Die Gleichgewichtskonstante einer Komplexierungsreaktion wird als Bildungskonstante Kf oder Stabilitätskonstante Ks bezeichnet.
Je höher der Wert der Bildungskonstante ist, desto größer ist die Stabilität des Komplexes.
Im Allgemeinen verlaufen die Komplexierungsreaktionen schrittweise, und die Gleichgewichtskonstante für jeden Schritt wird als stufenweise Bildungskonstante bezeichnet.
Für die gesamte Reaktion ist jedoch die kumulative oder Gesamtbildungskonstante β das Produkt aller schrittweisen Bildungskonstanten.
Im Gegensatz dazu ist die Dissoziationsreaktion des Komplexes das Gegenteil seiner Bildungsgleichgewichte, wobei die Gleichgewichtskonstante als Dissoziations- oder Instabilitätskonstante bezeichnet wird, ausgedrückt als Kehrwert der Bildungskonstante.

Komplexierungsreaktionen finden statt, wenn sich zwischen Metallionen und Liganden dative oder koordinative kovalente Bindungen bilden. Die bei diesen Reaktionen entstehenden Verbindungen werden Koordinationsverbindungen genannt. Die Anzahl der zwischen dem Metallion und den Liganden gebildeten Bindungen wird als Koordinationszahl bezeichnet. Im Allgemeinen werden die meisten Metallionen in einer wässrigen Lösung durch Wassermoleküle solvatisiert und liegen daher als Aquakomplexe vor.

Die Gleichgewichtskonstante der Komplexierungsreaktion wird als Bildungskonstante Kf, auch Stabilitätskonstante Ks genannt, dargestellt. Umgekehrt ist die Dissoziation des Komplexes die Umkehrung der Komplexbildung, und die Dissoziationsgleichgewichtskonstante wird Dissoziationskonstante Kd oder Instabilitätskonstante Ki genannt, die gleich dem Kehrwert der Bildungskonstante Kf ist.

Die Komplexierung erfolgt im Allgemeinen schrittweise, wobei ein Metallion zunächst mit einem Liganden und dann mit dem zweiten Liganden einen Komplex bildet, bis es seine Koordinationszahl erfüllt. Die Gleichgewichtskonstanten für jeden Reaktionsschritt sind die schrittweisen Bildungskonstanten, dargestellt als Kf1, Kf2, …, Kfn.. Die Gesamtformation oder kumulative Konstante β ist das Produkt der schrittweisen Bildungskonstanten oder einfach Kf.

Die Werte der schrittweisen Bildungskonstanten nehmen aufgrund statistischer, Coulomb- und sterischer Faktoren stetig ab. Statistische Faktoren kommen ins Spiel, nachdem sich der erste Ligand an das Metallion gebunden hat, was die Anzahl der verfügbaren Stellen für den nächsten Liganden und damit die Wahrscheinlichkeit der Bindung des nächsten Liganden verringert. Coulombische Faktoren spielen auch eine Rolle, wenn das Metallion mit dem ersten Liganden einen Komplex bildet, da die positive Ladung des Metallions abnimmt und somit die Coulomb-Anziehung für den nächsten Liganden abnimmt. Sterische Faktoren wirken sich am stärksten aus, wenn ein Ligand sperrig ist, da die von diesem Liganden erzeugten sterischen Abstoßungen die Leichtigkeit beeinflussen, mit der die nachfolgenden Liganden mit dem Metallion interagieren.

Stepwise metal-ligand complexation forms coordination compounds when metal ions and ligands share electrons through dative, or coordinate covalent, bonds. In these reactions, the metal ion binds to one or more ligands until its coordination number is reached. This number tells how many bonds form between the metal ion and the ligands.

In aqueous solution, many metal ions are already surrounded by water molecules. These species are called aqua complexes. Complexation reactions are described by the formation constant Kf, which is also called the stability constant Ks. The reverse process is complex dissociation, and its equilibrium constant is the dissociation constant Kd, or instability constant Ki, which equals the reciprocal of Kf.

Complexation usually occurs in stages. A metal ion first binds one ligand, then another, and continues step by step until the coordination number is satisfied. The equilibrium constant for each stage is written as Kf1, Kf2, and so on through Kfn. The overall formation constant, also called the cumulative constant β, is the product of the stepwise formation constants and is often written as Kf.

The stepwise formation constants usually decrease from one step to the next. This drop happens for statistical, coulombic, and steric reasons. Statistical factors matter because each ligand that binds leaves fewer open sites for the next one. Coulombic factors matter because the metal ion becomes less positively charged after binding, so it attracts the next ligand less strongly. Steric factors matter most when a ligand is bulky, since crowding and repulsion make later bindings harder.
