Treadmill Gait Measurement

Treadmill gait measurement is the quantitative assessment of locomotor behavior as an animal or person walks or runs on a motorized belt under controlled conditions. By setting belt speed and recording movement with video, force sensors, or limb-tracking systems, researchers can measure parameters such as stride length, cadence, stance and swing duration, symmetry, and step-to-step variability. These measurements provide objective indicators of motor coordination, balance, and adaptation to changing locomotor demands. In behavioral research, treadmill gait analysis helps characterize movement phenotypes, evaluate the effects of neurological injury or treatment, and compare locomotor performance across experimental conditions.

Here, we present a protocol to analyze the posture and the gait of patients after lumbar fusion surgery by means of high-resolution video rasterstereography and a treadmill equipped with an integrated sensor mat. Allowing critical functional postoperative evaluation on a less subjective level may enhance accuracy and reliability of indication for surgery.

Kinematic gait analysis in the sagittal plane yields highly precise information about how movement is executed. We describe the application of these techniques to identify gait deficits for mice subjected to autoimmune-mediated demyelination. These methods may also be used to characterize gait deficits for other mouse models featuring impaired locomotion.

Using a Split-belt Treadmill to Evaluate Generalization of Human Locomotor Adaptation

We describe a protocol for investigating human locomotor adaptation using the split-belt treadmill, which has two belts that can drive each leg at a different speed. We specifically focus on a paradigm designed to test the generalization of adapted locomotor patterns to different walking contexts (e.g., gait speeds, walking environments).

The Treadmill Fatigue Test: A Simple, High-throughput Assay of Fatigue-like Behavior for the Mouse

Fatigue is a common, undertreated and frequently poorly-understood symptom in many diseases and disorders. New preclinical assays of fatigue may help to improve current understanding and future treatment of fatigue. To that end, the current protocol provides a novel means of measuring fatigue-like behavior in the mouse.

Actin Treadmilling

2023

Actin filaments undergo polymerization and depolymerization from either end. The polymerization and depolymerization rates depend on the cytosolic concentration of free G-actins. The polymerization rate is generally higher at the plus or barbed end, while the depolymerization rate is higher at the minus or pointed end. At a steady state, critical concentration describes the concentration of free G-actin monomers at which the polymerization rate at the plus end is equal to that of the...
