Inguinal Adipose Tissue

Inguinal adipose tissue is a subcutaneous fat depot located near the groin and lower abdominal region, where adipocytes store energy and participate in whole-body metabolic regulation. Its cells accumulate triglycerides in lipid droplets and, in response to hormonal or sympathetic signals, activate lipolysis to release fatty acids; cold exposure and β-adrenergic stimulation can also promote the conversion of white adipocytes toward thermogenic beige cells through mitochondrial uncoupling protein 1 activity. In biochemistry and metabolic research, this depot helps investigators study adipocyte differentiation, energy balance, thermogenesis, insulin sensitivity, and obesity-related tissue remodeling.

Measurement of Basal and Forskolin-stimulated Lipolysis in Inguinal Adipose Fat Pads

This protocol describes the method of determining basal and forskolin-stimulated lipolysis in inguinal fat pads obtained from normal chow diet (NCD) or high fat diet (HFD) ± capsaicin fed wild type mice. As an index for lipolysis, glycerol release was measured from inguinal adipose fat pads.

Intravital Microscopy of the Inguinal Lymph Node

A technique for performing intravital microscopy of the inguinal lymph node (LN) is outlined. Such technique allows for real-time, in vivo study of the lymph node microvasculature and structure both during homeostasis and infection. This technique can be adapted to cell trafficking studies and to other lymph node sites.

Inguinal Subcutaneous White Adipose Tissue (ISWAT) Transplantation Model of Murine Islets

In this protocol, a method of murine islet isolation and transplantation into the inguinal subcutaneous white adipose tissue is described. Isolated syngeneic murine islets are transplanted into a murine recipient using a basement membrane hydrogel. The blood glucose level of the recipients is monitored, and histology analysis of the islet grafts is performed.

Adipose tissue (AT) is a site of intense immune cell activation and interaction. Almost all cells of the immune system are present in AT and their ratios are altered by obesity. Proper isolation, quantification, and characterization of AT immune cell populations are critical for understanding their role in immunometabolic disease.
