Plant Seedling Cultivation

Plant seedling cultivation is the controlled germination and early growth of plants from seeds, providing uniform biological material for studying development and metabolism. Seeds receive suitable moisture, temperature, oxygen, light, and, when needed, mineral nutrients so that imbibition activates enzymes, stored reserves are mobilized, and embryonic tissues develop into roots and shoots. In biochemistry, cultivated seedlings support analysis of enzyme activity, carbohydrate and protein metabolism, hormone signaling, and responses to environmental stress. Standardized growth conditions improve experimental reproducibility and help researchers connect biochemical changes with visible traits such as germination rate, biomass, and root architecture.

The described hydroponic cocultivation system supports intact plants with metal mesh screens and cocultivates them with bacteria. Plant tissue, bacteria, and secreted molecules can then be separately harvested for downstream analyses, simultaneously allowing for the molecular responses of both plant hosts and interacting microbes or microbiomes to be investigated.

This video shows a procedure for isolating intact protoplasts from tissues of 14-day-old seedlings of Arabidopsis. Given that the isolated protoplasts remain intact for at least 96h and are isolated from seedlings instead of one-month-old mature plants, this procedure expedites assays requiring intact protoplasts.
