Colony Pcr Screening

Colony PCR screening is a rapid method for identifying bacterial or yeast colonies that carry a desired DNA construct after transformation, making it useful for selecting candidate clones in bioengineering. A small amount of cells from each colony is used directly as the PCR template, while primers flanking an insert or spanning a junction amplify only colonies with the expected sequence; the products are then evaluated by gel electrophoresis. This approach can verify insert presence and approximate size before plasmid isolation and sequencing. By reducing time and reagent use during cloning workflows, colony PCR screening supports recombinant DNA assembly, strain engineering, and construction of libraries for downstream experiments.

In this video, we perform a clonogenic assay of keratinocytes harvested from adult mice to determine the ability of individual cells to proliferate and form colonies. This keratinocyte harvesting technique is useful for studying other downstream aspects of cutaneous chemical carcinogenesis and tumor promotion.

ScanLag: High-throughput Quantification of Colony Growth and Lag Time

ScanLag is a high-throughput method for measuring the delay in growth, namely lag time, as well as the growth rate of colonies for thousands of cells in parallel. Screening using ScanLag enables the discrimination between long lag-time and slow growth at the level of a single variant.
