Embryo Perfusion

Embryo perfusion is the controlled movement of a culture medium or fluid around and through an embryo or embryonic tissue to support development under laboratory conditions. By regulating flow, oxygen, nutrients, temperature, and waste removal, perfusion systems recreate aspects of the transport environment normally provided by the maternal and extraembryonic circulation. In bioengineering, these platforms help researchers maintain viable embryos, study how fluid transport and mechanical forces influence growth, and evaluate developmental responses to changing environmental conditions. They also support the design of microfluidic culture devices and engineered models that improve control, observation, and reproducibility in developmental research.

Antegrade Perfusion Model: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.

The porcine model of liver normothermic machine perfusion (NMP), described here, can be successfully used to study NMP as a preservation strategy, a tool for viability assessment, and a platform for organ repair.
