Ficoll 400

Ficoll 400 is a highly branched, water-soluble polymer derived from sucrose, with an average molecular mass of approximately 400 kDa, used to control solution density and macromolecular crowding. In density-gradient centrifugation, Ficoll 400 forms a viscous, osmotically compatible medium in which particles and cells migrate according to their size and buoyant density. Bioengineers use it to isolate cells, organelles, and biomolecular complexes, while its crowding effects can also model concentrated intracellular environments and influence molecular interactions. These properties support cell processing, tissue engineering research, biomolecule purification, and the design of more physiologically relevant in vitro systems.

This protocol describes a detailed method for efficient generation of integration-free iPSCs from human adult peripheral blood cells. With the use of four oriP/EBNA-based episomal vectors to express the reprogramming factors, KLF4, MYC, BCL-XL, or OCT4 and SOX2, thousands of iPSC colonies can be obtained from 1 mL of peripheral blood.
