Muscle Construct

A muscle construct is an engineered three-dimensional tissue that organizes muscle cells and supporting materials to reproduce key structural and functional features of native muscle. In bioengineering, cells are embedded in or aligned within a scaffold or hydrogel, where biochemical cues, mechanical loading, and electrical stimulation can promote differentiation, maturation, and coordinated contraction. These constructs provide controlled models for studying muscle development, injury, disease, and drug responses while reducing reliance on animal experiments. They also support regenerative medicine research by informing the design of implantable tissues and strategies to restore strength and function after muscle damage.

Here, we demonstrate fabrication of collagen-based, tissue constructs containing skeletal myoblasts. These 3-D engineered constructs may be used to replace or repair tissues in vivo. For our purposes, we have designed these as an atrioventricular electrical conduit for the repair of complete heart block[1].

We describe a method to directly measure muscle force, muscle power, contractile kinetics and fatigability of isolated skeletal muscles in an in vitro system using field stimulation. Valuable information on Ca2+ handling properties and contractile machinery of the muscle can be obtained using different stimulating protocols.

This paper describes the assembly process and operation of a bench-scale photosynthetic bioreactor that can be used, in conjunction with other methods, to estimate pertinent kinetic growth parameters. This system continuously monitors the pH, light, and temperature using sensors, a data acquisition and control unit, and open-source data acquisition software.
