Scaffold Pore Size

Scaffold pore size is the characteristic diameter of voids within a three-dimensional biomaterial, an important design parameter in bioengineering and tissue engineering. By controlling fluid movement, nutrient and oxygen diffusion, waste removal, and cell migration through interconnected pores, pore size influences how cells attach, proliferate, and organize within the scaffold; larger pores can improve transport and infiltration but may reduce surface area and mechanical strength. Optimizing pore size helps balance biological accessibility with structural support for applications such as bone, cartilage, and soft-tissue regeneration, while guiding scaffold fabrication and the development of functional tissue substitutes.

In this work, poly(lactic acid)/polyethylene glycol (PLA/PEG) scaffolds were prepared by using a combination of melt mixing and selective leaching. The method herein discussed permitted to develop three-layer scaffolds by highly controlling both porosity and pore size. The mechanical properties were also evaluated in a physiological environment.

We present two processes for the microfabrication of porous polymer chips for three-dimensional cell cultivation. The first one is hot embossing combined with a solvent vapour welding process. The second one uses a recently developed microthermoforming process combined with ion track technology leading to a significant simplification of manufacture.

This video describes the technique of seeding mesenchymal stem cells (MSCs) onto a biodegradable poly(lactic acid) (PLA) scaffold. The prepared scaffolds can be implanted into the surgical resection cavity of the brain from where they may migrate towards tumor foci. Further, these cells can be engineered to express therapeutic proteins to stimulate apoptosis in cancer cells.
