Spheroid Production

Spheroid production is the generation of three-dimensional, multicellular aggregates that mimic key features of tissue organization more closely than conventional two-dimensional cultures. In bioengineering, cells are encouraged to self-assemble under nonadherent conditions, such as low-attachment plates or hanging drops, where cell-cell adhesion and compaction form a dense spheroid. Researchers can control spheroid size and composition by adjusting cell number, culture conditions, and time, while monitoring viability, structure, and growth. These models support tissue engineering, drug screening, disease modeling, and studies of cell behavior, providing physiologically relevant platforms for evaluating cell interactions and improving engineered tissues.

An Efficient and Flexible Cell Aggregation Method for 3D Spheroid Production

Here, we describe a rapid and flexible protocol for the formation of 3D cell spheroids through cell aggregation. This is easily adapted to multiple cell types and is suitable for use in a variety of applications including cell migration, invasion, or anoikis assays, and for imaging and quantifying cell-matrix interactions.

Herein, we communicate protocols to efficiently generate and administer therapeutic spherical aggregates or 'spheroids' of MSCs primed under xeno-free conditions for experimental and clinical applications.
