Tissue Specimen Preparation

Tissue specimen preparation is the process of preserving, handling, and processing biological tissues so their structure and composition can be examined reliably. It typically involves fixation to stabilize cellular components, followed by dehydration, embedding, sectioning, and staining or other labeling methods that produce interpretable specimens for microscopy and analysis. In bioengineering, these procedures support the evaluation of engineered tissues, biomaterials, implants, and tissue–device interfaces by revealing morphology, cell distribution, and material integration. Consistent preparation reduces structural artifacts and improves the accuracy, reproducibility, and scientific value of histological and imaging-based assessments.

Cryo Electron Microscopes, either Scanning (SEM) or Transmission (TEM), are widely used for characterization of biological samples or other materials with a high water content1. A SEM/Focused Ion Beam (FIB) is used to identify features of interest in samples and extract a thin, electron-transparent lamella for transfer to a cryo-TEM.
