Transillumination Imaging

Transillumination imaging is a noninvasive optical technique that forms images by illuminating a specimen and recording light transmitted through it, revealing internal structure without cutting or staining. As light travels through tissue or another turbid material, absorption and scattering alter its intensity and spatial distribution; a camera or sensor converts these variations into contrast that reflects differences in composition, thickness, or density. In bioengineering, the method supports visualization of soft tissues, microvascular features, embryos, and engineered constructs, particularly when rapid, label-free assessment is useful. Its simple optical setup can complement microscopy and help monitor structure and function during development or fabrication.

Mesoscopic fluorescence tomography operates beyond the penetration limits of tissue-sectioning fluorescence microscopy. The technique is based on multi-projection illumination and a photon transport description. We demonstrate in-vivo whole-body 3D visualization of the morphogenesis of GFP-expressing wing imaginal discs in Drosophila melanogaster.

Reporter Enzyme Fluorescence Imaging for In Vivo Tracking of Bacterial Infection

This video demonstrates the reporter enzyme fluorescence (REF) imaging for in vivo detection of Mycobacterium tuberculosis infection. The infected mice are intraperitoneally injected with a substrate for REF imaging. Upon being hydrolyzed by bacterial β-lactamase enzyme, the substrate emits fluorescence, the detection of which aids evaluation of infection.

We describe a protocol for hybrid imaging, combining fluorescence-mediated tomography (FMT) with micro computed tomography (µCT). After fusion and reconstruction, we perform interactive organ segmentation to extract quantitative measurements of the fluorescence distribution.
