Mitochondrial Succinate Dehydrogenase

Mitochondrial succinate dehydrogenase is an enzyme complex in the inner mitochondrial membrane that links the tricarboxylic acid cycle with the electron transport chain. It oxidizes succinate to fumarate, transferring the released electrons to flavin adenine dinucleotide and then through iron-sulfur centers to ubiquinone, contributing to proton-driven ATP production without directly pumping protons. Measuring succinate dehydrogenase activity through spectrophotometric assays, histochemical staining, or respirometry helps assess mitochondrial abundance and function. These approaches support studies of cellular metabolism, tissue pathology, inherited mitochondrial disorders, and metabolic changes associated with cancer or other disease states.

The cytochrome c oxidase/sodium dehydrogenase (COX/SDH) double-labeling method allows for direct visualization of mitochondrial respiratory enzyme deficiencies in fresh-frozen tissue sections. This is a straightforward histochemical technique and is useful in investigating mitochondrial diseases, aging, and aging-related disorders.

Simultaneous Measurement of Superoxide/Hydrogen Peroxide and NADH Production by Flavin-containing Mitochondrial Dehydrogenases

Mitochondria contain several flavin-dependent enzymes that can produce reactive oxygen species (ROS). Monitoring ROS release from individual sites in mitochondria is challenging due to unwanted side reactions. We present an easy, inexpensive method for direct assessment of native rates for ROS release using purified flavoenzymes and microplate fluorometry.

Animal Mitochondrial Genetics

2020

Among all the organelles in an animal cell, only mitochondria have their own independent genomes. Animal mitochondrial DNA is a double-stranded, closed-circular molecule with around 20,000 base pairs. Mitochondrial DNA is unique in that one of its two strands, the heavy, or H, -strand is guanine rich, whereas the complementary strand is cytosine rich and called the light, or L, -strand. Compared to nuclear DNA, mitochondrial DNA has a very low percentage of non-coding regions and is marked by...

A Lactate Dehydrogenase Release Assay To Detect Macrophage Death Following Nigericin Exposure

2025

This video demonstrates the lactate dehydrogenase assay to detect inflammasome-mediated macrophage death. The macrophages are exposed to lipopolysaccharide for priming, followed by exposure to nigericin to cause inflammasome-induced cell death, which is determined by the lactate dehydrogenase assay.

A Lactate Dehydrogenase Release Assay to Detect Cryptococcus neoformans Infection in Macrophages
