Lipid Staining

Lipid staining is a group of analytical and histological techniques that makes lipids visible in cells, tissues, or biological samples, helping researchers assess their location, abundance, and organization. These methods use dyes or labeling reagents that preferentially dissolve in, bind to, or react with hydrophobic lipid domains, producing a detectable color or fluorescence under appropriate imaging conditions. In biological techniques, lipid staining supports microscopy-based studies of lipid droplets, membranes, adipose tissue, and pathological accumulation, while enabling comparisons among developmental states, experimental treatments, or disease models. Careful sample preparation and stain selection are essential because different lipids and preservation conditions yield different signals.

In this video, we demonstrate in vitro staining of neutral lipids in hepatocytes to detect hepatic steatosis or excessive fat accumulation in liver. Excessive fat is accumulated in the cells in form of lipid droplets. Oil Red O is a lipid soluble dye that binds to intracellular neutral lipids in these lipid droplets. When observed under a light microscope, these intracellular lipid droplets appear intensely red in color.
