Mycolic Acid Separation

Mycolic acid separation is the isolation and resolution of long-chain fatty acids from complex biological lipid mixtures, a process important for studying the distinctive cell envelopes of mycobacteria. Typically, chemical extraction or saponification releases these highly hydrophobic molecules, after which thin-layer chromatography or liquid chromatography separates them according to polarity, chain structure, and interactions with the stationary phase. The resulting profiles support mycobacterial identification and chemotaxonomy, while purified mycolic acids enable analysis of cell-wall architecture, biosynthesis, host interactions, and drug targets. These techniques also help compare lipid composition across species, strains, and experimental conditions.

Isotachophoresis (ITP) is a robust electrokinetic separation and preconcentration technique with applications ranging from toxin detection to sample preparation. We review the physical principles of ITP and the methodology of applying this technique to two specific example applications: separation and detection of small molecules and purification of nucleic acids from cell culture lysate.
