Hybrid Separation Method

A hybrid separation method is a biological separation strategy that combines two or more distinct separation principles to isolate, purify, or characterize cells, organelles, or biomolecules more effectively than a single method alone. It works by applying complementary steps, such as differences in size, density, charge, or molecular affinity, so that one process reduces sample complexity and a subsequent process improves selectivity or resolution. In biological techniques, hybrid separation methods support sample preparation, protein and nucleic acid purification, cell fractionation, and analytical workflows, helping researchers obtain cleaner, more defined material for downstream measurement, diagnosis, or experimentation.

Here, we present a simple method for performing fluorescence DNA in situ hybridization (DNA ISH) to visualize repetitive heterochromatic sequences on slide-mounted chromosomes. The method requires minimal reagents and it is versatile for use with short or long probes, different tissues, and detection with fluorescence or non-fluorescence-based signals.
