Protein Phospholipid Interactions

Protein phospholipid interactions are associations between proteins and the lipid molecules that form biological membranes, influencing membrane structure, signaling, and transport. They arise through complementary electrostatic and hydrophobic forces, hydrogen bonding, and specific recognition of lipid headgroups or acyl chains, with membrane composition and charge affecting binding strength and orientation. Biological techniques such as lipid-binding assays, membrane reconstitution, spectroscopy, and microscopy help characterize these interactions under controlled conditions. Their analysis supports research on membrane protein function, vesicle trafficking, signal transduction, and drug targeting, while revealing how lipid environments regulate protein activity and cellular organization.

Quantitative Flow Cytometry to Study Labeled Protein-Phospholipid Vesicle Interactions

In this video, we demonstrate the interaction between proteins and artificial phospholipid vesicles using quantitative flow cytometry. The binding of fluorescently-labeled proteins to fluorescently-labeled phospholipid vesicles increases the mean fluorescence intensity, and this increase is detected by a flow cytometer.
