Adenovirus Transduction

Adenovirus transduction is a gene-delivery technique that uses adenoviral vectors to introduce DNA into target cells, enabling controlled study or production of gene expression. The vector binds cell-surface receptors, enters by receptor-mediated endocytosis, escapes the endosome, and transports its genetic payload to the nucleus, where the DNA typically remains episomal rather than integrating into the host genome. In biology, this approach supports transient or high-level expression in cultured cells and tissues for functional genomics, protein production, vaccine development, and gene therapy research; however, vector design, cell type, and host immune responses influence efficiency and duration.

Source: Chen, H., et al. Live Cell Imaging of the TGF-β/Smad3 Signaling Pathway In Vitro and In Vivo Using an Adenovirus Reporter System. J. Vis. Exp. (2018).This video demonstrates a method to study SMAD pathway activation in breast cancer cells using dual adenoviral infection. One virus delivers a constitutively expressed GFP gene, while the other carries an RFP reporter under a SMAD-sensitive promoter. Upon TGF-beta stimulation, SMAD proteins are activated, forming a complex that induces RFP...

Here we describe a protocol based only on cell infection, which improves the efficiency of recombinant parvovirus production by more than 100 fold in comparison to other protocols in use. This protocol relies on the use of a novel adenovirus 5-based helper containing the parvovirus VP transcription unit (Ad-VP).
