Cartilage Regeneration

Cartilage regeneration is the biological repair or replacement of damaged cartilage, a resilient connective tissue that cushions joints and supports smooth movement. Because mature cartilage lacks blood vessels and contains relatively few chondrocytes, it has limited intrinsic healing capacity; regeneration strategies therefore aim to supply cells, structural scaffolds, and biochemical signals that promote extracellular-matrix production and restore tissue organization. In biology and regenerative medicine, approaches such as microfracture, autologous cell transplantation, biomaterial-based tissue engineering, and stem-cell therapies are used to investigate repair of articular cartilage after injury or degenerative disease. Understanding these mechanisms may improve joint function and guide development of durable, patient-specific treatments.

3D Magnetic Stem Cell Aggregation and Bioreactor Maturation for Cartilage Regeneration

Chondrogenesis from stem cells requires fine tuning the culture conditions. Here, we present a magnetic approach for condensing cells, an essential step to initiate chondrogenesis. In addition, we show that dynamic maturation in a bioreactor applies mechanical stimulation to the cellular constructs and enhances cartilaginous extracellular matrix production.

This protocol describes the assembly of a layer-by-layer Janus base nano-matrix (JBNm) scaffold by adding Janus base nanotubes (JBNts), matrilin-3, and Transforming Growth Factor Beta-1 (TGF-β1) sequentially. The JBNm was fabricated and characterized; additionally, it displayed excellent bioactivity, encouraging cell functions such as adhesion, proliferation, and differentiation.
