Chemical Gradient

A chemical gradient is a spatial change in the concentration of a substance, creating directional information that guides biological activity. Molecules move down their concentration gradient by diffusion, while cells detect local differences through receptors and adjust movement, growth, gene expression, or developmental fate. In biology, gradients of nutrients, signaling molecules, and morphogens help direct chemotaxis, establish tissue patterns, and coordinate communication between cells. Studying chemical gradients clarifies how organisms organize themselves, respond to changing environments, and develop from individual cells into complex tissues, with applications in developmental biology, microbiology, tissue engineering, and regenerative research.

This paper reports a polydimethylsiloxane-polycarbonate (PDMS-PC) microfluidic device capable of reliably generating combinations of chemical and oxygen gradients for cell migration studies, which can be practically utilized in biological labs without sophisticated instrumentation.

We describe a protocol for the microfabrication of the gradient-generating microfluidic device that can generate spatial and temporal gradients in well-defined microenvironment. In this approach, the gradient-generating microfluidic device can be used to study directed cell migration, embryogenesis, wound healing, and cancer metastasis.
