Dig Labeled Rna Probe

A DIG-labeled RNA probe is a nonradioactive, sequence-specific nucleic acid tool used to detect complementary RNA or DNA molecules in biological samples. During probe synthesis, digoxigenin (DIG) is incorporated into the RNA, and the labeled probe hybridizes to a complementary target sequence under controlled temperature and salt conditions. After washing removes unbound probe, an anti-DIG antibody linked to an enzyme or fluorescent reporter produces a detectable signal. DIG-labeled RNA probes support in situ hybridization, gene expression analysis, and transcript localization in cells, tissues, and embryos, providing sensitive alternatives to radioactive probes for developmental biology and molecular research.

Here a method to localize bacteria within paraffin-embedded tissues using DIG-labeled 16S rRNA-targeting DNA probes has been described. This protocol can be applied to study the role of bacteria in various diseases such as periodontitis, cancers, and inflammatory immune diseases.
