Epithelial Elongation

Epithelial elongation is the lengthening of epithelial cells or tissue along a defined axis, a shape change that helps organize and remodel biological structures. It can occur when polarized actomyosin contractility reshapes individual cells, or when neighboring cells intercalate, narrowing tissue in one direction while extending it in another; adherens junctions coordinate these movements and preserve tissue cohesion. In biology, epithelial elongation contributes to embryonic morphogenesis, organ formation, and epithelial repair after injury. Studying this process clarifies how cell shape, mechanical forces, and tissue architecture interact, with implications for understanding developmental disorders and abnormal tissue remodeling.

Here we detail protocols specifically designed to monitor morphogenic defects that occur during early and late phases of embryonic elongation of the nematode Caenorhabditis elegans. Ultimately, these protocols are designed to identify genes that regulate these phases and to characterize their differential requirements along the antero-posterior axis of the embryo.

We present here a cell culture method for inducing mesenchymal-epithelial transitions (MET) in sarcoma cells based on combined ectopic expression of microRNA-200 family members and grainyhead-like 2 (GRHL2).
