Ex Vivo Podocyte Model

An ex vivo podocyte model is a laboratory system that maintains podocytes, the specialized cells that help form the kidney’s glomerular filtration barrier, outside the organism for direct study. Researchers isolate podocytes or intact glomerular tissue and sustain it under controlled conditions, allowing examination of cell morphology, cytoskeletal organization, barrier function, and responses to defined stimuli while preserving aspects of the native renal environment. In biology, these models support investigation of podocyte injury and proteinuria, including changes linked to genetic, inflammatory, or drug-related stress. They also provide a useful platform for testing disease mechanisms and evaluating potential therapeutic interventions before animal or clinical studies.

Changes in the intracellular calcium levels in the podocytes are one of the most important means to control the filtration function of glomeruli. Here we explain a high-throughput approach that allows detection of real-time calcium handling and single ion channels activity in the podocytes of the freshly isolated glomeruli.

Here, we present a systematic approach for developing physiologically relevant, sensitive and specific in vivo assays for interpreting variation in human pathology. Transient genetic manipulation via microinjection of WT and mutant human mRNA and morpholino (MO) antisense oligonucleotides harness the tractability of the developing zebrafish embryo to rapidly assay pathogenic mutations, especially, but not exclusively, in the context of human developmental disorders.

This manuscript describes the development of an animal model that allows for the direct testing of the effects of tumor hypoxia on metastasis and the deciphering the mechanisms of its action. Although the experiments described here focus on Ewing sarcoma, a similar approach can be applied to other tumor types.
