Fast Green Dye

Fast Green dye is a water-soluble biological stain used to visualize proteins and cellular structures, making otherwise transparent specimens easier to examine by light microscopy. As an acidic triphenylmethane dye, it binds primarily to basic regions of proteins through electrostatic interactions, producing a green signal whose intensity depends on dye concentration, staining time, and tissue composition. In biology, Fast Green supports histological staining of cytoplasm, connective tissue, and other protein-rich components, and it can complement counterstains in protocols such as trichrome staining. These applications help researchers distinguish tissue organization, assess cellular morphology, and interpret structural changes in experimental samples.

Measuring Fast Calcium Fluxes in Cardiomyocytes

We present a method to isolate rapid (microsecond) calcium events from slower fluxes in living cells using laser scanning confocal microscopy. The method measures fluorescence intensity fluctuations of calcium indicators by recording line scans of several hundred pixels in a cell. Histogram analysis allows us to isolate the time scales of different calcium fluxes.
