Fly Diet Manipulation

Fly diet manipulation is the controlled adjustment of food composition or availability to examine how nutrition influences physiology, behavior, development, and lifespan in flies. In laboratory studies, researchers vary nutrients such as carbohydrates, proteins, lipids, vitamins, or total calories, then monitor how these changes alter energy balance, metabolism, reproduction, or stress responses. Using standardized diets and controlled feeding conditions makes it possible to separate nutritional effects from genetic and environmental factors. This approach, especially in Drosophila research, provides a practical model for investigating metabolic regulation, aging, disease biology, and the biological consequences of nutrient imbalance.

Rearing the Fruit Fly Drosophila melanogaster Under Axenic and Gnotobiotic Conditions

A method for rearing Drosophila melanogaster under axenic and gnotobiotic conditions is presented. Fly embryos are dechorionated in sodium hypochlorite, transferred aseptically to sterile diet, and reared in closed containers. Inoculating diet and embryos with bacteria leads to gnotobiotic associations, and bacterial presence is confirmed by plating whole-body Drosophila homogenates.

High Fat Diet Feeding and High Throughput Triacylglyceride Assay in Drosophila Melanogaster

This is a high fat diet feeding protocol to induce obesity in Drosophila, a model for understanding fundamental molecular mechanisms implicated in lipotoxicity. It also provides a high throughput triacylglyceride assay for measuring fat accumulation in Drosophila and potentially other (insect) models under various dietary, environmental, genetic or physiological conditions.

Experimental Manipulation of Body Size to Estimate Morphological Scaling Relationships in Drosophila

Morphological scaling relationships capture and describe organismal shape. We present a method to measure morphological scaling relationships across the natural range of body sizes in fully metamorphic insects. Using a simple diet manipulation we increase the distribution of trait sizes, permitting the accurate description of how shape and size co-vary.
