Graded Ethanol

Graded ethanol is a sequence of ethanol solutions with progressively changing concentrations, used to control dehydration and rehydration in biological sample preparation. By moving a specimen through incremental ethanol concentrations rather than transferring it directly between water and absolute ethanol, the process gradually replaces water in cells and tissues while reducing osmotic and structural stress. This controlled transition supports tissue processing, histology, microscopy, and other workflows that require samples to be dehydrated before embedding, clearing, or imaging. The chosen concentration steps and exposure times influence morphology preservation, reagent penetration, and the quality and reproducibility of biological observations.

Microdialysis of Ethanol During Operant Ethanol Self-administration and Ethanol Determination by Gas Chromatography

A method to determine the time course of ethanol concentration in the brains of rats during operant ethanol self-administration is described. Gas chromatography with flame ionization detection is used to quantify ethanol in the dialysate samples, because it has the sensitivity required for the small volumes that are generated.
