Heterozygous Mice

Heterozygous mice are animals that carry two different alleles of a gene, one inherited from each parent, making them valuable models for studying how genetic variation influences biological traits. In these mice, a functional allele may partially or fully compensate for a disrupted, deleted, or disease-associated allele, depending on gene dosage and the resulting protein activity. Researchers use heterozygous mice to examine dominant and recessive inheritance, assess gene function, and model conditions in which a single altered copy affects physiology or development. Comparing heterozygous animals with homozygous and wild-type controls can clarify genetic mechanisms and support the development of disease models and therapeutic strategies.

Behavioral testing is used in pre-clinical trials to assess the phenotypic effects of a disease or treatment on an animal's wellbeing. In order to globally assess motor functioning, we selected tests for general locomotion, muscular strength, and coordination: the open field test, the mesh test, and the rotarod test, respectively.

We describe the isolation of cardiac extracellular matrix from C57Bl/6J control mice, tight-skin mice, and tight-skin mice treated with the IRF5 inhibitory peptide. We also describe the vasodilation studies on the isolated vessels from C57Bl/6J, tight-skin mice and tight-skin mice treated with the IRF5 inhibitory peptide.

Novel strategies to faithfully model somatic mutations in hematopoietic stem and progenitor cells (HSPCs) are necessary to better study hematopoietic stem cell biology and hematological malignancies. Here, a protocol to model heterozygous gain-of-function mutations in HSPCs by combining the use of CRISPR/Cas9 and dual rAAV donor transduction is described.

This article describes a new protocol for the quantitative measurement of social motivation in mice using operant conditioning for a social reward. The protocol is useful for the measurement of social motivation in mouse models of autism and other disorders of social behavior.
