Intercellular Junctions

Intercellular junctions are specialized structures that connect neighboring cells, enabling tissues to maintain strength, selective barriers, and coordinated activity. They work through transmembrane adhesion proteins that link adjacent cells directly or connect cell membranes to the cytoskeleton; some junctions also form channels for intercellular communication. Tight junctions regulate movement between cells, adherens junctions and desmosomes provide mechanical support, and gap junctions allow ions and small molecules to pass between cells. Studying these junctions helps explain tissue organization, epithelial barrier function, development, and diseases involving disrupted adhesion or cell signaling.

Nanopodia are thin but fragile membrane channels that extend up to 100 μm from a cell's leading front or trailing rear and sense the cellular environment. Direct fixation at 37 °C, gentle washing, and avoidance of organic solvents like ethanol, methanol, or acetone and of higher Triton X-100 concentrations are required to observe these cellular structures.

Intercellular Communication in a Contact-Free Co-Culture of Neurons and Oligodendrocytes

Begin with a contact-free co-culture of neurons and oligodendrocytes.A permeable membrane separates the neurons in the transwell insert from the oligodendrocytes at the bottom well.Add potassium to the insert to raise extracellular potassium levels, which inhibits the efflux of intracellular potassium. The potassium accumulation depolarizes the neurons, triggering the opening of voltage-gated calcium channels and allowing calcium influx. The influx stimulates the release of excitatory...

This protocol describes a scalpel loading-fluorescent dye transfer technique that measures intercellular communication through gap junction channels. Gap junctional intercellular communication is a major cellular process by which tissue homeostasis is maintained and disruption of this cell signaling has adverse health effects.

A p-n junction is formed when p-type and n-type semiconductor materials are joined together. At the interface of the p-n junction, holes from the p-side and electrons from the n-side begin to diffuse into the opposite sides due to the concentration gradient. This diffusion of carriers leads to a region around the junction where there are no free charge carriers, known as the depletion region. The charge density within the depletion region for the n-side and p-side can be described by the...
