Multi-well Plate Assay

A multi-well plate assay is a laboratory method that analyzes many biological samples or reactions simultaneously in separate wells of a standardized plate, improving experimental throughput and consistency. Each well receives a defined sample, reagent, or treatment, and an assay-specific reaction produces a measurable optical, fluorescent, luminescent, or other signal that is compared with controls or standards. In biology, these assays support measurements of enzyme activity, cell viability, protein binding, gene expression, and responses to drugs or environmental conditions. Their parallel format helps researchers conserve samples and reagents while generating quantitative data for screening, comparison, and statistical analysis.

Systematic Analysis of In Vitro Cell Rolling Using a Multi-well Plate Microfluidic System

This study used a multi-well plate microfluidic system, significantly increasing throughput of cell rolling studies under physiologically relevant shear flow. Given the importance of cell rolling in the multi-step cell homing cascade and the importance of cell homing following systemic delivery of exogenous populations of cells in patients, this system offers potential as a screening platform to improve cell-based therapy.

In this video, we demonstrate an assay that investigates monoamine release mechanisms from the neurons of metabolically active ex vivo rat brain slices, when treated with test drugs. The assay demonstrates that neurostimulant drugs induce monoamine release via the cellular monoamine transporters, whereas high KCl concentration induces synaptic vesicle exocytosis-mediated vesicular monoamine release.
