Obligate Symbionts

Obligate symbionts are organisms that depend on a close, long-term relationship with a host or partner for survival, growth, or reproduction. Their dependence often results from metabolic integration: one organism supplies essential nutrients or cellular functions that the other cannot produce independently, while maintaining specialized structures or signaling systems for exchange. In biology, obligate symbionts include intracellular bacteria, fungi associated with plants, and animal-associated microbes, and their partnerships can be mutualistic, parasitic, or context-dependent. Studying these relationships clarifies coevolution, host physiology, and ecosystem function, with applications in microbiome research, crop improvement, disease biology, and the development of targeted interventions.

Single-plant, Sterile Microcosms for Nodulation and Growth of the Legume Plant Medicago truncatula with the Rhizobial Symbiont Sinorhizobium meliloti

Growth of Medicago truncatula plants in symbiosis with the nitrogen-fixing bacteria Sinorhizobium meliloti in individual, sterile microcosms made from standard laboratory plates permits frequent examination of root systems and nodules without compromising sterility. Plants can be maintained in these growth chambers for up to 9 weeks.

Transposon-insertion Sequencing as a Tool to Elucidate Bacterial Colonization Factors in a Burkholderia gladioli Symbiont of Lagria villosa Beetles

This is an adapted method for identifying candidate insect colonization factors in a Burkholderia beneficial symbiont. The beetle host is infected with a random mutant library generated via transposon mutagenesis, and library complexity after colonization is compared to a control grown in vitro.

Methods for the Extraction of Endosymbionts from the Whitefly Bemisia tabaci

Here, we present a protocol to isolate endosymbionts from the whitefly Bemisia tabaci through dissection and filtration. After amplification, the DNA samples are suitable for subsequent sequencing and study of the mutualism between endosymbionts and whitefly.
