Ocular Tissue Homogenization

Ocular tissue homogenization is the controlled disruption of eye tissues to produce a uniform sample for biological and biochemical analysis. Mechanical force, often applied with a homogenizer or grinding device, breaks tissue architecture and releases cells, proteins, nucleic acids, lipids, and other intracellular components into a homogenate while preserving the analytes needed for downstream assays. Researchers use this method to measure gene and protein expression, analyze enzymes and metabolites, and investigate disease-related changes in structures such as the retina, cornea, lens, and optic nerve. Careful control of tissue handling, temperature, and homogenization intensity improves sample consistency and supports reliable comparisons in ocular biology.

We describe a technique for the preparation of clarified human cortical homogenates, protein separation by SDS-PAGE, antigen retrieval and immunoblotting with an antibody to the Aβ peptide. Using this protocol, we consistently detect monomeric and multimeric Aβ in cortical tissue from humans with Alzheimer's pathology.
