Parathyroid Principal Cells

Parathyroid principal cells, also called chief cells, are the hormone-producing cells of the parathyroid glands and central regulators of blood calcium homeostasis. They monitor extracellular calcium through the calcium-sensing receptor (CaSR); when calcium levels fall, signaling promotes synthesis and release of parathyroid hormone (PTH), whereas elevated calcium activates feedback that suppresses secretion. PTH then acts on bone and kidneys, indirectly supporting intestinal calcium absorption, to restore calcium balance and coordinate phosphate regulation. Studying these cells helps explain normal endocrine physiology and disorders such as hyperparathyroidism, while informing research into calcium-related diseases and parathyroid function.

Two Techniques to Create Hypoparathyroid Mice: Parathyroidectomy Using GFP Glands and Diphtheria-Toxin-Mediated Parathyroid Ablation

Mice with acquired hypoparathyroidism would be useful for studying novel drug therapies for hypoparathyroidism. Two procedures to create such mice are demonstrated. The GFP-PTX mouse is generated by surgical parathyroidectomy guided by green fluorescing parathyroid glands. A second, non-surgical approach is based on parathyroid-specific expression of the diphtheria toxin receptor.
