Phage Dna Recombination

Phage DNA recombination is the exchange or rearrangement of genetic material in bacteriophages, viruses that infect bacteria, and it is central to phage replication, genome maintenance, and evolution. During infection, recombination can occur when homologous DNA sequences pair and exchange strands through strand invasion and branch migration, or when specialized enzymes join DNA at defined sites; host and phage repair pathways can help resolve the resulting structures. These processes generate genetic diversity, restore damaged genomes, and support the transfer of genetic traits. Studying phage DNA recombination helps researchers map genes, engineer phage genomes, understand virus-host interactions, and develop tools for biotechnology and antibacterial research.

2014

Phage display is a powerful technique to capture proteins or protein moieties that interact with an immobilized molecule of interest. Once a decision of the type of phage cDNA library to create and screen has been made, the protocol described here permits efficient affinity selection leading to identification of interactors.

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.

2021

Scientists create recombinant DNA by combining DNA from different sources—often, other species—in the laboratory. DNA cloning allows researchers to study specific genes by inserting them into easily manipulated cells, such as bacteria. Organisms that contain recombinant DNA are known as genetically modified organisms (GMOs). Recombinant DNA technology produces organisms with new genes that can benefit science, medicine, and agriculture. How Do Scientists Create Recombinant DNA?Creation of...

Source: Eggers, C. H. Phage-Mediated Genetic Manipulation of the Lyme Disease Spirochete Borrelia burgdorferi. J. Vis. Exp. (2022).This video demonstrates the isolation of borrelial phage particles from a Borrelia burgdorferi suspension using polyethylene glycol-mediated phase separation. The sample is centrifuged, treated with a salt solution, and a molecular crowding agent to precipitate phages. Chloroform extraction is done to remove debris and concentrate phage particles. The purified phage...
