Photon Flux Measurement

Photon flux measurement quantifies the number of photons reaching a surface or detector per unit time, providing a basis for assessing light intensity in biological systems. Instruments such as photodiodes, quantum sensors, and integrating spheres convert incident photons into an electrical signal or photon count, often reporting photon flux density as photons per square meter per second across a defined wavelength range. In biology, these measurements characterize illumination for photosynthesis, plant growth, photobiology experiments, and light-controlled cell or tissue studies. We present a method to isolate rapid (microsecond) calcium events from slower fluxes in living cells using laser scanning confocal microscopy. The method measures fluorescence intensity fluctuations of calcium indicators by recording line scans of several hundred pixels in a cell. Histogram analysis allows us to isolate the time scales of different calcium fluxes.
