Porcine Oocyte Isolation

Porcine oocyte isolation is the laboratory process of recovering egg cells from pig ovarian tissue, providing material for studying reproduction, embryology, developmental biology, and assisted reproductive technologies. In a typical workflow, follicles are aspirated or opened to release cumulus-oocyte complexes, which are collected, washed, and selected under a microscope according to features such as intact cumulus layers and uniform cytoplasm. Isolated oocytes can then undergo in vitro maturation, fertilization, embryo culture, cryopreservation, or molecular analysis, making this technique important for investigating oocyte quality, early development, reproductive biotechnology, and porcine models of human disease.

We present simple techniques for isolating giant transcriptionally active lampbrush chromosomes from living oocytes of frogs and salamanders. We describe how to observe these chromosomes "alive" by phase contrast or differential interference contrast, and how to fix them for fluorescent in situ hybridization or immunofluorescent staining.
