Pyruvate Dehydrogenase

Pyruvate dehydrogenase is a multienzyme complex that converts pyruvate, the end product of glycolysis, into acetyl-CoA, linking carbohydrate breakdown to the citric acid cycle. Through oxidative decarboxylation, the complex removes carbon dioxide from pyruvate, transfers the remaining two-carbon fragment to coenzyme A, and produces NADH using coordinated cofactors including thiamine pyrophosphate, lipoamide, FAD, and NAD+. Its activity is regulated by phosphorylation and cellular energy status, helping balance fuel use and biosynthetic demands. Studying pyruvate dehydrogenase clarifies how cells generate energy, integrate metabolic pathways, and respond to disruptions in mitochondrial metabolism.
